WB Use a concentration of 1 µg/ml. Predicted molecular weight: 17 kDa. Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.

    Monothiol glutaredoxin involved in the biogenesis of iron-sulfur clusters. Required for normal iron homeostasis. Required for normal regulation of hemoglobin synthesis by the iron-sulfur protein ACO1.
  • Involvement in disease
    Defects in GLRX5 are a cause of anemia sideroblastic pyridoxine-refractory autosomal recessive (PRARSA) [MIM:205950]. A form of sideroblastic anemia not responsive to pyridoxine. Sideroblastic anemia is characterized by anemia of varying severity, hypochromic peripheral erythrocytes, systemic iron overload secondary to chronic ineffective erythropoiesis, and the presence of bone marrow ringed sideroblasts. Sideroblasts are characterized by iron-loaded mitochondria clustered around the nucleus.

Following up my message from yesterday, I at least confirmed with our source for this antibody that the immunogen is unique to GLRX5:

"Based on the BLAST results using the sequence of the immunizing peptide, it is uniquely 100% homologous to the human GLRX5 protein. In fact, the next highest homologous human protein is ATP5B with 50% homology, with no other glutaredoxins listed."

They also report: "We do not have a published reference where someone has used this antibody to detect endogenous GLRX5 protein."

If your results are impossible to interpret because of the multiple bands you are seeing, I recommend isolating a mitochondrial fraction of your samples, to enrich for GLRX5, and hopefully diminish the signal from the other proteins that the antibody is cross-reacting with. Your colleague and I discussed some issues your lab was having with Abcam's Glutaredoxin 5 antibody, ab86411. I am waiting for the lab to send any additional testing we have, which I will forward.
