  • ab85137 staining S100 in a human melanoma cell line by Flow Cytometry. The cells were harvested using EDTA and washed in PBS. The sample was incubated with the primary antibody (1/100 in PBS) for 15 minutes at room temperature. A DyLight® 488-conjugated goat anti-mouse IgG H&L (ab96879) (1/100) was used as the secondary antibody.

  • Overlay histogram showing Jurkat cells stained with ab8090 (red line). The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab8090, 0.01μg/1x106 cells) for 30 min at 22°C. The secondary antibody Goat anti-mouse IgG H&L (DyLight® 488, pre-adsorbed) (ab96879) was used at 1/2000 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 0.01μg/1x106 cells) used under the same conditions. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.

    The cells were fixed with 4% paraformaldehyde (10 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab2861, 1µg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) ab96879 at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab91361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed.

  • Emission spectra of DyLight® fluorochromes available in our catalog.
    Line colors represent the approximate visible colors of the wavelength maxima.

  • ICC/IF image of ab40084 stained HepG2 cells. The cells were 4% PFA fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab40084, 5µg/ml) overnight at +4°C. The secondary antibody (green) was DyLight® 488 goat anti-mouse IgG - H&L, pre-adsorbed (ab96879) used at a 1/250 dilution for 1h. Alexa Fluor® 594 WGA was used to label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

Regarding PPAR-delta ab:
I think ab58137 (www.abcam.com/ab58137) as you suggested would be the best antibody to try as it has been tested in flow cyt and using the same secondary ab (ab96879) would be fine. However you would have to use a mouse IgG1 as a control as the primary antibody (ab58137) has the isotype IgG1. I suggest isotype control antibody, mouse IgG1 [ICIGG1] ab91353 (www.abcam.com/ab91353).
Regarding PPAR-gamma ab, we do not have any antibodies which have been tested in flow cytometry.

You are correct, you need a secondary antibody that is conjugated to a fluorescent molecule (or an enzyme, followed by the enzyme's substrate to get some color) in order to see where ab9489 is sticking to CD31. The one you picked out, ab96879, is a good choice, assuming you have a microscope that detects fluorescence and has the correct filter for that fluorochrome, DyLight® 488. (It very likely does).

ab96876 is a Donkey polyclonal anti mouse IgG secondary, and should therefore detect ab13579TRF2 antibody [4A794]which is a mouse IgG1 primary.

As stated on the datasheet, ab96876 has been tested in IHC-P, ICC/IF, Flow Cyt, and WB.

It is therefore tested and guaranteed for western blot and immunohistochemistry on paraffin embedded sections. I am sorry I am not sure what you mean by IHC both FL - Non FL, could you clarify?

The fluorochromes which are sold under the brand names Alexa, DyLight or Chromeo are designed to ensure an optimal performance and thus are chemically almost identical. As you may have noticed the advantagesfor these include excellent photostability and bright fluorescence (see also flyer attached). So from this point of view, there is not much of a difference between the DyLights or Chromeos.

Therefore, you may want to make your decision based on other factors:

We offer DyLight conjugated antibodies in the wavelengths 488nm, 550nm, 594nm and 650nm (based on the excitation maxima). These fluorochromes are suitable for most settings. We have Chromeo conjugated antibodies in comparable wavelengths available (488nm, 546nm, 642nm), but also for 494nm and 505nm which are somewhere in between. So if you wish to stain your target in ones of these colours (and your microscope has the correct filter settings), I would recommend the Chromeos.

On the other hand, our DyLight antibody range contains a wide variety of antibody formats, such as antibodies against H&L chain (e.g. ab96879), against the Fc part (e.g. ab98736) or F(ab')2 fragments (e.g. ab98739). Also, within our DyLight antibody range we have a number of secondaries available which have been pre-absorbed against other species to minimise potential cross-reactivity (e.g. ab96879). These advantages might be relevant for you depending on the sample type and species and the primary antibody.

Regarding the resistance to photobleaching, I would slightly prefer the DyLight conjugated antibodies, asaccording to customer feedback they seem to perform excellent. However, this is my opinion only and merelybased on our sales volume for the DyLights; we do not have any further data to confirm this.
