    Human Eotaxin-2 ELISA Kit (ab226898) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Eotaxin-2 protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Eotaxin-2 with 15.35 pg/ml sensitivity.


    SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:


            - Single-wash protocol reduces assay time to 90 minutes or less
            - High sensitivity, specificity and reproducibility from superior antibodies
            - Fully validated in biological samples
            - 96-wells plate breakable into 12 x 8 wells strips


    A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

  • Notes

    Eotaxin-2, also known as CCL24, MPIF-2 or Ckß-6, is a C-C chemokine originally identified in activated monocytes. Eotaxin-2 is expressed in activated T lymphocytes, GM-CSF treated macrophages and dermal fibroblasts. Secretion of Eotaxin-2 is known to recruit and activate CCR3-bearing cells particularly eosinophils as well as basophils. Activation of eosinophils generates reactive oxygen species, lipid mediators of inflammation and degranulation of toxic granule proteins. Chemotaxis and activation of basophils, previously primed with IL-3, lead to release of histamine and leukotriene C4.

      The concentrations of Eotaxin-2 were measured in duplicates, interpolated from the Eotaxin-2 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 50%, plasma (citrate) 50%, plasma (EDTA) 3.12%, and plasma (heparin) 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Eotaxin-2 concentration was determined to be 13,298 pg/mL in serum, 10,364 pg/mL in plasma (citrate), 9,354 pg/mL in plasma (EDTA), and 8,191 pg/mL in plasma (heparin).

      PBMCs were cultured in RPMI media and either stimulated with 1.5% PHA or unstimulated for 48 hours. The concentrations of Eotaxin-2 were measured in duplicate and interpolated from the Eotaxin-2 standard curve and corrected for sample dilution. Undiluted samples are as follows: PBMC stimulated 3.12% and PBMC unstimulated 50%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Eotaxin-2 concentration was determined to be 107,776 pg/mL in stimulated PBMCs and 677 pg/mL in unstimulated PBMCs.
