    Abcam’s FGF basic Mouse ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of mouse FGF basic in serum, plasma and cell culture supernatants.

    This assay employs an antibody specific for mouse FGF basic coated on a 96-well plate. Standards and samples are pipetted into the wells and FGF basic present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse FGF basic antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of FGF basic bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

I assume you are looking for a lysis buffer because you wish to test cell or tissue lysates samples with the kit? If so, please note that the ab100670 kit has not been validated with lysate samples, only serum, plasma, and cell culture supernatants. If you would like to try and test this kit with lysates, attached is a file which gives guidelines to preparing a compatible lysis buffer. This buffer is very similar to a RIPA buffer.

Although this kit was neither designed nor tested with cell or tissue lysate samples, this kit as well as our other sandwich ELISA kits are very accommodating to many different sample types so lysates samples should work.

There are actually some publications available, successfully testing tissue lysates with this kit.

Thank you for contacting us. We do not currently sell any ELISA kits that are specifically designed for use on rat samples. After checking with the lab, our mouse and human ELISA kits for these targets will be unlikely to cross-react with rat, but have not been tested on rat samples.

We do currently sell antibodies for these targets that would be suitable for use in an ELISA on rat samples:

FGF basic: no antibodies tested for ELISA on rat

Unfortunately, epitope mapping of the two antibody pairs has not been performed, so we do not know the exact amino acid sequence recognised and if these regions are specific for human and mouse, respectively. All we know for the antibody pair in the mouse FGF ELISA kit is that the antibody pair is binding on the whole mature protein between amino acids x and y.

Due to the high degree of conservation of basic FGF the mouse FGF ELISA Kit ab100670 will indeed also recognise human FGF2 and vice versa, as internal testing data also show.
