  • Relevance
    IgG is a monomeric immunoglobulin, built of two heavy chains gamma and two light chains. Each molecule has two antigen binding sites. This is the most abundant immunoglobulin and is approximately equally distributed in blood and in tissue liquids, constituting 75% of serum immunoglobulins in humans. This is the only isotype that can pass through the human placenta, thereby providing protection to the fetus in its first weeks of life before its own immune system has developed. It can bind to many kinds of pathogens, for example viruses, bacteria, and fungi, and protects the body against them by complement activation (classic pathway), opsonization for phagocytosis and neutralisation of their toxins. There are 4 subclasses: IgG1 (66%), IgG2 (23%), IgG3 (7%) and IgG4 (4%).

  • The wells were coated with ab195576 at 0.5µg/ml at 50µl/well overnight at 4°C, followed by a 5% BSA blocking step for 2h RT. Human IgG (ab98981) was then added starting at 40 µg/ml and plasma/serum at 1:50 and gradually diluted 1:4, 50µl/well for 2h. ab201484 was then added at 1:10,000 dilution, 50µl/well for 2h. A HRP-streptavidin (ab7403) was used at 1:10,000 dilution for 1h.

These two IgG proteins are similar in many ways including origin, and levels of purity. However they do differ in regards to the storage buffer in which they are provided. ab98981: Preservative: None Constituents: 0.02M Ammonium bicarbonate (May contain traces of buffer salts) ab91102: Preservative: 0.05% Sodium Azide Constituents: 20mM PBS, 150mM Sodium chloride, pH 7.4. The Anti-Lambda Light chain [HP6054] antibody, ab168861, will react with any Lambda light chain which is present in these products and is guaranteed to work in WB, IHC-P, IHC-Fr. In reviewing the IgG proteins I do not see that these are kappa or lambda Light chain. The total kappa to lambda ratio is roughly 2:1 in serum and I would expect a similar ratio with these products.

I would recommend use of a secondary antibody against mouse IgG which has been pre-absorbed against human IgG. These antibodies are run through columns containing IgGs of many species and and antibody which reacts with those IgGs will be trapped in the columns. I cannot see why the Human IgG protein (ab98981) could not be used in cell culture. However, I would bear in mind that the product has not been sterilized and may contaminate the cell culture and as we have not used it ourselves in this application ourselves we would not be able to guarantee its performance.

These two products are from different sources and there are slight variations between them.

ab91102 is from one patient, tested in SDS-PAGE and the storage buffer contains 0.05% Sodium Azide, 20mM PBS, 150mM Sodium chloride, pH 7.4.

The other product ab98981 is purified from pooled normal Human serum/plasma, tested in RID and SDS-PAGE and the storage buffer contains 0.02M Ammonium bicarbonate (no preservative).

The storage at 2-8 is relevant to the freeze dried product. Once resuspended we would not really recommend storage at 2-8 for no more than 1-2 weeks, and this would depend on the diluent used. We would suggest aliquoting and storing frozen at <-15, preferable -80C for long term storage.

But I can confirm that ab7500 is guaranteed for peptide ELISA with human samples by our Abpromise guarantee.

Thank you for your enquiry. The denaturing process is reversible in some cases. The original structure of some proteins can be regenerated by removal of the denaturing agent.
