      The ESAT6 antigen from Mycobacterium tuberculosis is a dominant target for cell mediated immunity in the early phase of tuberculosis (TB) in TB patients as well as in various animal models. ESAT6 is not found in M. bovis BCG. Its function is not known but it elicits high level of IFN-gamma from memory effector cells during the first phase of a protective immune response.

    The efficiency of ESAT-6 as a stimulant of T-cells for IFN-g production.

    The aim of this research was to test the ability of ESAT-6 to stimulate T-cell production of Interferon-gamma ,IFN-g.
    Blood from a consenting QuantiFERON positive participant was collected. Monocytes, CD4+ and CD8+ cells were isolated from the participant’s blood. The monocytes served as antigen presenting cells, therefore 100 000 monocytes were co-cultured with 100 000 CD4+ or CD8+ cells respectively. Thus, each well had a total of 200 000 cells and a final volume of 200ul.
    These co-cultures were then stimulated according to set conditions. Some cells were left unstimulated to serve as a negative control. Recombinant ESAT-6 ,ab124574 - Recombinant Mycobacterium tuberculosis ESAT6 protein from Abcam, was used to stimulate CD4+ and CD8+ cells at concentrations of 2.5ug/ml, 5ug/ml or 10ug/ml. Phytohemagglutinin -PHA- 10ug/ml, and BCG ,MOI 5:1, were used as positive control stimulants. The stimulated cultures were incubated for 72 hours at 37 C and 5% CO2.
    After 72 hours the supernatants were collected from each culture well. The IFN-γ concentration from each culture was quantified with Human IFN-g Quantikine ELISA. The results were expressed in a scatter plot depicting the concentration of IFN-g ,pg/ml, secreted by CD4+ and CD8+ cells in response to each stimulation condition ,Figure 1.
    The results show an increase in IFN-g production for all stimulation conditions compared to the unstimulated cultures. Upon comparing ESAT-6 at different concentrations, ESAT-6 at 10ug/ml induced the highest IFN-γ secretion for both CD4+ and CD8+ cells.
    It can be concluded that ESAT-6 can be used as a stimulant in a stimulation assay to activate T cells to produce IFN-g.
