Transcription factor for the thyroid-specific expression of the genes exclusively expressed in the thyroid cell type, maintaining the functional differentiation of such cells. -
Tissue specificity
Expressed in the excretory system, thyroid gland and Wilms tumors. -
Involvement in disease
Defects in PAX8 are the cause of congenital hypothyroidism non-goitrous type 2 (CHNG2) [MIM:218700]. CHNG2 is a disease characterized by thyroid dysgenesis, the most frequent cause of congenital hypothyroidism, accounting for 85% of case. The thyroid gland can be completely absent (athyreosis), ectopically located and/or severely hypoplastic. Ectopic thyroid gland is the most frequent malformation, with thyroid tissue being found most often at the base of the tongue. -

ChIC/CUT&RUN was performed using a pAG-MNase at a final concentration of 700 ng/µL, 2.5 x 10^5 NIH:OVCAR-3 (Human ovary adenocarcinoma epithelial cell) cells and 5 µg of ab181054 [EPR13510]. The resulting DNA was sequenced on the Illumina NovaSeq 6000 to a depth of 10 million reads. The negative IgG control ab172730 is also shown. The University of Geneva owns patents relevant to ChIC (Chromatin Immuno-Cleavage) methods.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) analysis of human thyroid tissue labelling PAX8 with purified ab181054 at a dilution of 1/1000. Heat mediated antigen retrieval was performed using Tris/EDTA buffer pH 9. ab97051, a HRP-conjugated goat anti-rabbit IgG (H+L) was used as the secondary antibody (1/500). Negative control using PBS instead of primary antibody. Counterstained with hematoxylin.
