Target

  • Function
May act in negative regulation of cell growth and transformation by interacting with nonreceptor tyrosine kinases ABL1 and/or ABL2. May play a role in regulation of EGF-induced Erk pathway activation. Involved in cytoskeletal reorganization and EGFR signaling. Together with EPS8 participates in transduction of signals from Ras to Rac. In vitro, a trimeric complex of ABI1, EPS8 and SOS1 exhibits Rac specific guanine nucleotide exchange factor (GEF) activity and ABI1 seems to act as an adapter in the complex. Regulates ABL1/c-Abl-mediated phosphorylation of ENAH. Recruits WASF1 to lamellipodia and there seems to regulate WASF1 protein level.
  • Tissue specificity
Widely expressed, with highest expression in brain.
  • Involvement in disease
Note=A chromosomal aberration involving ABI1 is a cause of acute leukemias. Translocation t(10;11)(p11.2;q23) with MLL. ABI1 isoform 2 was found to be present in acute leukemia MLL-ABI1 fusion transcript.
  • Sequence similarities
    Belongs to the ABI family.
    Contains 1 SH3 domain.
    Contains 1 t-SNARE coiled-coil homology domain.
  • Domain
The t-SNARE coiled-coil homology domain is necessary and sufficient for interaction with STX1A.
  • Post-translational
    modifications
In vitro substrate for v-Abl (By similarity). Phosphorylated on tyrosine residues after serum stimulation or induction by v-Abl.
  • Cellular localization
Cytoplasm. Nucleus. Cell projection > lamellipodium. Cell projection > filopodium. Cell projection > growth cone. Cell junction > synapse > synaptosome. Cytoplasm > cytoskeleton. Localized to protruding lamellipodia and filopodia tips. Also localized to neuronal growth cones and synaptosomes.

Images

  • IHC image of ab11222 staining in human normal skin formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab11222, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Answer

Thank you for your telephone call this morning. I am sorry to hear you have had difficulty obtaining satisfactory results from this antibody.

I would like to reassure you that this antibody is tested and covered by our 6 month guarantee forWB and for mouse and human samples. In the event that a product is not functioning in the tested applications and species cited on the product data sheet (and the problem has been reported within 6 months of purchase), we will be pleased to provide a credit note, free of charge replacement or refund.

As discussed on the telephone I would like to investigate this particular case further for you, and also obtain more information for our quality monitoring records. In order to proceed with this, I have enclosed a technical questionnaire below. I would appreciate if you could complete this. It will help you put the information we require together very easily.

I would appreciate if it is alsopossible to provide an image which would help us to assess the results.

Thank you for your time and cooperation. We look forward to receiving the completed questionaire.

Answer

Thank you for contacting us. For an isotype control, I recommend ab18469, mouse IgG2b. The concentration is 0.5 mg/ml, so the dilution should be half that of ab11222 (which has a stock concentration of 1.0 mg/ml) to match the concentrations of the working solutions. https://www.abcam.com/index.html?datasheet=18469 For a western blot positive control, I suggest a HeLa cell lysate, such as ab29545: https://www.abcam.com/index.html?datasheet=29545 I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for your enquiry. SSH3BP1 antibody [4E2] (ab11222) is a mouse monoclonal antibody. The secondary that you employ will depend on the approach that you use, whether an enzymatic approach or a flurophore approach. I would like to recommend that you hover on our product browser on our home page (www.abcam.com) and navigate to "secondaries", "anti-mouse", "anti-IgG" and then either select "enzymatic" or "flurophore". I hope this information helps, please do not hesitate to contact us if you need any more advice or information.

Answer

Thank you for your enquiry. SSH3BP1 antibody [4E2] (ab11222) was developed against the spectrin SH3 domain binding protein Hssh3bp1 (Ziemnicka-Kotula et al., 1998). Hssh3bp1 is a marker of macropinocytic vesicles and a potential regulator of macropinocytosis (Xu et al., 2000). The gene encoding Hssh3bp1 has a role as a tumor suppressor gene which is inactivated during prostate tumorigenesis. (Macoska et al., 2001). I hope this information helps, please do not hesitate to contact us if you need any more advice or information.
