Properties

Applications

Our Abpromise guarantee covers the use of ab13677 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 0.01 - 0.1 µg/ml. Detects a band of approximately 45 kDa (predicted molecular weight: 48 kDa).Can be blocked with Human ACADM / MCAD peptide (ab23229).
IHC-P Use a concentration of 3 - 5 µg/ml. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.

Target

  • Function
This enzyme is specific for acyl chain lengths of 4 to 16.
  • Pathway
Lipid metabolism; mitochondrial fatty acid beta-oxidation.
  • Involvement in disease
Defects in ACADM are the cause of acyl-CoA dehydrogenase medium-chain deficiency (ACADMD) [MIM:201450]. It is an autosomal recessive disease which causes fasting hypoglycemia, hepatic dysfunction, and encephalopathy, often resulting in death in infancy.
  • Sequence similarities
Belongs to the acyl-CoA dehydrogenase family.
  • Cellular localization
    Mitochondrion matrix.

    Developed using the ECL technique.

    Predicted band size: 48 kDa

Ab13677 staining (0.05µg/ml) of Human Heart lysate (RIPA buffer, 35µg total protein per lane).  Primary incubated for 1 hour.  Detected by western blot using chemiluminescence.

    Primary incubated for 1 hour.

  • ab13677 (3.8 µg/ml) staining ACADM / MCAD in paraffin-embedded human Liver. Steamed antigen retieval with citrate buffer (pH 6) was carried out.
