I would ideally like a replacement antibody as I still need to validate the ADAM12 result for publication. Would you be able to advise whether you think any of the alternative ADAM12 antibodies would be suitable for a flow cytometry application. Looking at the data sheets, although none of your alternative antibodies are tested in flow cytometry, both ab115203 and ab28747 are supposed to recognise a N-terminal fragment of the protein and give a band by western blotting of the correct size. However I think they both possibly recognise a region within the signal peptide sequence of ADAM12 rather than the rest of the protein – is this corrent. AB56366 is also supposed to recognise N-terminal amino acids 208-305, which I believe should also be extracellular, however it has only been used of recombinant fragments of hADAM12, rather than endogenous protein. Do you have any other data on these antibodies to suggest that they may be useful for flow cytometry?
