  • Relevance

    ADCY10 belongs to a distinct class of mammalian adenylyl cyclase that is soluble and insensitive to G protein or forskolin regulation. It is localized in the cytoplasm and is thought to function as a general bicarbonate sensor throughout the body. It may also play an important role in the generation of cAMP in spermatozoa, implying possible roles in sperm maturation through the epididymis, capacitation, hypermotility, and/or the acrosome reaction.

Please find the answers to the questions below:

1.) I suggest to use 10-30ug of total protein per well on the gel. This might have to be optimised, but I recommend to start with 20ug and optimise form there.

The samples are heated at 95C form 10 minutes.

2.) 10 x PBS

Dissolve the following in 800ml distilled H2O.
80g of NaCl

2.0g of KCl

14.4g of Na2HPO4

2.4g of KH2PO4

Adjust pH to 7.4.

Adjust volume to 1L with additional distilled H2O.

Sterilize by autoclaving.

For a 1X solution, mix 1 part of the 10X solution with 9 parts distilled waterand pH to 7.4 again.

3.) PBS - Tween was used for washing:

1 x PBS with 0.1% Tween (For 1 liter: 100 ml of PBS 10X + 900 ml distilled water + 1ml Tween-20)
Usually there are 3 washes after each incubation time that are 5 minutes long each.

4.) I can recommend to use the same blocking agent in all solutions and not to mix different blocking agents. For the antibody solutions 5% blocking agnet can be used, but I recommend to start with 1% blocking agent and to increase this only when high background occurs.
