WB Use a concentration of 1 µg/ml. Detects a band of approximately 22 kDa (predicted molecular weight: 20 kDa).Can be blocked with Human Adrenomedullin/ADM peptide (ab69116).
IHC-P Use a concentration of 5 µg/ml.

Target

  • Function

    AM and PAMP are potent hypotensive and vasodilatator agents. Numerous actions have been reported most related to the physiologic control of fluid and electrolyte homeostasis. In the kidney, am is diuretic and natriuretic, and both am and pamp inhibit aldosterone secretion by direct adrenal actions. In pituitary gland, both peptides at physiologically relevant doses inhibit basal ACTH secretion. Both peptides appear to act in brain and pituitary gland to facilitate the loss of plasma volume, actions which complement their hypotensive effects in blood vessels.

    Exposure time: 8 minutes
  • IHC image of Adrenomedullin/ADM staining in Human placenta formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab69117, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

    For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

We have not carried out a peptide blocking study for this product. This has been characterised using WB against MCF7, DU145 and MDA MB 361, and IHC against Human plancenta.


We guarantee the antibody to be specific for adrenomedullin for 12 months after purchase, so if you determine that it is not specific in the tested applications and species listed on the datasheet we will refund or replace the product free of charge.

Answer

I can confirm that this antibody has been tested for both applications Western blot (WB) and IHC on formalin-fixed paraffin-embedded tissue sections (IHC-P) using human samples. The cross-reactivity with other species has not been determined yet, though ab69117 is expected to react with mouse, rat, horse and pig Adrenomedullin due to sequence homology.

Please find my answers below:

1) could you tell me to which exact residues the antibody was raised against?

A: The exact immunogen sequence is commercially sensitive information and I can only release that the synthetic peptide conjugated to KLH is derived from within residues 1 - 100 of Human Adrenomedullin.



2) has your antibody been tested against human, mouse, rat and in both IHC and Western Blotting?

If so could you send me images for each of the results you obtained in these tests?

A: The antibody has only been tested in human samples in WB and IHC-P as the datasheet indicates.



3) would it be possible to get an test sample of your antibody for me to test it in these application in my systems?

A: Since mouse, rat are only predicted species, we could offer you a testing discount.
