  • Relevance

Aggrecan is a member of a family of large, aggregating proteoglycans (also including versican, brevican and neurocan) which is found in articular cartilage. Aggrecan is composed of three major domains: G1, G2, and G3. Between the G1 and G2 domains there is an interglobulin region (IGD). The IGD region is the major site of cleavage by specific proteases like metalloproteinases (MMPs) and aggrecanase. Aggrecan cleavage has been associated with a number of degenerative diseases including rheumatoid arthritis and osteoarthritis. There is evidence that this family of proteoglycans modulates cell adhesion, migration, and axonal outgrowth in the CNS.

Thank you for your reply.

I can confirm that Anti-Chondroitin Sulfate antibody [CS-56], (ab11570) has been raised against the chicken protein. I am sorry this antibody has been sources externally, and regrettably we are not able to obtain the chicken sequence in order to align this with the sheep. The sheep sequence also appears to be unavailable.

For antigen retrieval, there has been an Abreview submitted by another customer who has used a commercially available antigen retrieval solution(Trilogy). I can recommend that antigen retrieval will often require some optimization. You may like to consider trying citrate bufferretrieval pH 6with several time points (3, 5, 10 and 20 minutes) to start with.

We do sell some pre prepared antigen retreival solutions:

I am sincerely sorry we have no more information regardingpotential crossreactivity with sheep again on this occasion. If you have any further questions, please do not hesitaet to contact us.

Thank you for contacting us and for your interest in our products.

I am sorry to confirm that as far as we are aware, ab118610has never been tested with samples from sheep. All tested and guaranteedspecies cross-reactivity information is stated on our datasheets, and these are updated as soon as any new information is brought to our attention.

The exact immunogen sequence is proprietary in this case. However, I can tell you thatit is located between amino acids 2100 and 2150 of NP_037359.3. I was hoping to do an alignment of the sheep protein with the imunogen for you. However, I am sorry I am regrettablynot able to find the sheep sequence of the Aggrecan protein in the protein databases. If you have a copy of this, I can recommend to try the alignment. We recommend alignment should be over 85% to predict that an antibody will detect in a different species. Alternatively, I am happy totry this alignment for you if you are able to send the sheep sequence.

If the alignment is high and you would like to test the antibody in this untested species, please contact me by replying to this message prior to the purchase as you may be eligible for our testing discount program.

Otherwise, we like to encourage all of our customers to submit an Abreview via the online product datasheet. We always appreciate customer feedback, whether positive or negative, and we make all product information available to researchers. Each Abreview earns Abpoints that can be used for discounts on future purchases or rewards such as Amazon.com gift certificates.

Thank you for contacting us. We need more information on the use of ab11570 and ab8610 in alphaLISA. Could you please answer the following questions? - Are you going to use these antibodies together or in combination with other antibodies? - Are you planning to conjugate these antibodies with different acceptor r donor molecules? - Are you planning to use these antibodies with different conjugated antibodies? e.g acceptor or donor molecule conjugated antibodies. - These antibodies are not tested together in Sandwich ELISA so how you are planning to proceed with the procedure? I am looking forward to hearing from you soon.
