Sections were fixed in 4% formaldehyde for 30 min, rinsed with PBS and then incubated with blocking buffer (PBS, 10 % normal donkey serum; NDS) for 10 min. Sections were then incubated overnight at 4 degrees C with primary antibodies diluted in PBS containing 2% NDS. After additional rinsing in PBS, the coverslips were incubated in fluorescent-labeled secondary antibodies (Cy2-, Rhodamine Red-X-, or Cy5-labeled donkey IgG) in PBS with 2% NDS for 30 min at RT.

  • ab24343 staining ALDH1A1 - Neuronal Marker (green) in Mouse pancreatic neoplasia tissue sections by Immunohistochemistry (IHC-Fr - frozen sections). Tissue was fixed with acetone/methanol (1:1) and blocked with 10% serum for 1 hour at room temperature. Samples were incubated with primary antibody (1/100 in PBS) for 4 hours. An Alexa Fluor®488-conjugated Donkey anti-rabbit IgG polyclonal (1/1000) was used as the secondary antibody. Nuclei were stained by DAPI (blue).

We had orders from XXXXXXXXXXX in year 2008, the lot we sent was 369562. This lot is currently out of stock, so I am sorry we will not be able to send this lot. My suggestion would be to try a new vial of same lot http://it.adminsite.com/admin/stock/stock_list.cfm?intAbID=24343&intStockID=373998&fMain=1&fViewExpired=1&strViewOffice=%25 or trying a thin section would also help.

We generally recommend our customers trying 5-10 micron sections. I can recommend 8 micron thick sections than 16; the results will be much nicer. This could be reason why the antibody is not giving high intensity staining.

I have searched the order history of this antibody however I could not find any previous order from University of Amsterdam. The idea was to compare the current lot with previous one.

We unfortunately have same lot i.e. 373998 in stock; would you like to try the new vial of same lot?

We have not received any other complaint for this batch so the problem might be with the vial or with the protocol. In order to understand the problem better I would like to ask few questions;

- Are the sections paraffin embedded or frozen?
- Have you done any antigen retrieval?
- What tissue type was used for staining e.g. brain or liver
- Is the secondary antibody fine?
- Could you explain, how the tissue sections were prepared?
- Do you know the lot number which previously worked?
- In most cases; no staining improves by using high concentration of antibody. Have you tried any troubleshooting of protocol.

Please note because this is a rabbit polyclonal antibody so there will be some batch to batch variations; protocol optimizations would be highly recommended.

At this timeonepossible way toobtain the protein is likely toa obtain theplasmid expressing mouse ALDH1A1 in E. coli or some other vector, and then isolate the protein yourself.
