Our Abpromise guarantee covers the use of ab197531 in the following tested applications.
The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.
FunctionIs a component of the nuclear coiled bodies (CBS) which are involved in the function or assembly/disassembly of nucleoplasmic snRNPs. During mitosis, CBS disassemble, coinciding with a mitotic-specific phosphorylation of p80 coilin.
Tissue specificityFound in all the cell types examined.
Sequence similaritiesBelongs to the coilin family.
Cellular localizationNucleus. Nuclear coiled body located in the interchromatin space between the nucleolus and the nucleus.

Overlay histogram showing HeLa cells stained with ab197531 (red line). The cells were fixed with 80% methanol (5 min) and then permeabilized with 0.1% PBS-Tween for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab197531, 1/500 dilution) for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2b (monoclonal) Alexa Fluor® 488 (ab171465) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 20mW Argon ion laser (488nm) and 525/30 bandpass filter.
ab197531 staining Coilin in HeLa cells. The cells were fixed with 100% methanol (5min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab197531 at 1/1000 dilution (shown in green) and ab6046 (Rabbit polyclonal to beta Tubulin) at 1µg/ml. This was followed by an incubation at room temperature for 1h with ab150088, Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (Alexa Fluor® 594), pre-adsorbed, at 1µg/ml (shown in red). Nuclear DNA was labelled with DAPI (shown in blue).
