Target
-
Function
This is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of sodium and potassium ions across the plasma membrane. This action creates the electrochemical gradient of sodium and potassium ions, providing the energy for active transport of various nutrients. -
Sequence similarities
Belongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IIC subfamily. -
Post-translational
modificationsPhosphorylation on Tyr-10 modulates pumping activity. -
Cellular localization
Cell membrane. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV. - Information by UniProt

Immunocytochemistry - Alexa Fluor® 555 Anti-Sodium Potassium ATPase antibody [EP1845Y] - Plasma Membrane Loading Control (ab274883)
Immunofluorescence analysis of MCF7 (Human breast adenocarcinoma cell line) cells labeling Sodium Potassium ATPase with ab274883.
The cells were fixed with 100% methanol (5 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were tehn incubated with ab274883 at 1/100 dilution, overnight at +4°C (Green) and ab190573, Rabbit monoclonal to alphaTubulin (Alexa Fluor® 647), at 1/250 dilution (Red). Nuclear DNA was labelled with DAPI (Blue). Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).
