  • Function
    On ligand binding, forms a receptor complex consisting of two type II and two type I transmembrane serine/threonine kinases. Type II receptors phosphorylate and activate type I receptors which autophosphorylate, then bind and activate SMAD transcriptional regulators. Receptor for TGF-beta. May bind activin as well.
  • Involvement in disease
    Defects in ACVRL1 are the cause of hereditary hemorrhagic telangiectasia type 2 (HHT2) [MIM:600376]; also known as Osler-Rendu-Weber syndrome 2 (ORW2). HHT2 is an autosomal dominant multisystemic vascular dysplasia, characterized by recurrent epistaxis, muco-cutaneous telangiectases, gastro-intestinal hemorrhage, and pulmonary, cerebral and hepatic arteriovenous malformations; all secondary manifestations of the underlying vascular dysplasia.

  • ab51870 at 1/50 dilution staining ALK-1 in Human placental tissue section by Immunohistochemistry (Formalin/PFA fixed paraffin-embedded sections).

  • Overlay histogram showing JEG3 cells stained with ab51870 (red line). The cells were fixed with 80% methanol (5 min) and incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab51870, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG2a [ICIGG2A] (ab991361, 1µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in JEG3 cells fixed with 4% paraformaldehyde (10 min) used under the same conditions.

If you have a chance, it would be still worth running a good positive control (cytospin preparation from A549 cells or tissue section from placenta) which endogenously express TGF beta Receptor I. This product was launched in March 2006 so it has been on the market for several years.

There are 8 specific publications and the following papers have been used in immunostaining:

Therefore, any TGF beta Receptor Iantibody tested in the species and application you are using should be suitable for your experiments.

I can confirm that ab49575 clone [8A11] and ab51870 clone MM0015-8G33 have both been tested in IHC on paraffin embedded sections (IHC-P).

Although we produce many in-house antibodies, we obtain other antibodies from a wide range of sources. Therefore, comparison experiments will not often have been done.

If you wish to be more certain, I can suggest choosing the antibody that has been best characterized. By this I mean the antibody that has been tested in the most applications and has the most data available (on the datasheet).
