  • Undifferentiated human embryonic stem cells (H9 line) were stained with the tissue non-specific alkaline phosphatase antibody, ab17973. The cells were positive for alkaline phosphatase with staining localised on the surface membrane.

    Dilution used: 1/50

    Note that the feeder cells in the bottom left hand corner of the plate are not stained by this antibody.

  • ab17973 staining Alkaline Phosphatase, Tissue Non-Specific in human adrenal gland tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded tissue sections). The sections were fixed in paraformaldehye prior to blocking with 10% serum for 1 hour at 20°C. The primary antibody was diluted 1/500 and incubated with the sample for 12 hours at 4°C. An HRP-conjugated rabbit anti-mouse polyclonal was used as the secondary antibody, diluted 1/200.

The lab has tested ab17973 with H9 cells, NTERA-2 cl.D1 cells, iPSCs and A431 cells, while ab17272 has been tested with U2-OS cells. So, any of these cells can be used as positive controls.

Ab17973 was tested in the lab using the following protocol:

Cells were fixed using 4% paraformaldehyde and 2% sucrose in PBS for 20 min at room temperature and washed three times with PBS. The cells were then blocked overnight with a solution containing 5% donkey serum, 1% bovine serum albumin and 0.1% Triton X-100 in PBS. The samples were then washed six times using PBS and incubated with ab17973 (˜1:50-1:100 dilution) in the blocking solution without any detergents for at least 24 hours at 4°C. After incubation with the primary antibody, samples were washed six times (5 min each), and incubated in secondary antibody, for 30 min at room temperature, and washed with PBS six times (5 min each), stained with Hoechst 33285 and mounted on slides.

It is only known to react with the human Liver/Bone/Kidney isozyme of Alkaline Phosphatase (AP). Also, this antibody does not crossreact with other isoforms of alkaline phosphatase.
