Target

  • Function

This is the catalytic component of the active enzyme, which catalyzes the hydrolysis of ATP coupled with the exchange of sodium and potassium ions across the plasma membrane. This action creates the electrochemical gradient of sodium and potassium ions, providing the energy for active transport of various nutrients.
  • Sequence similarities

Belongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IIC subfamily.
  • Post-translational
    modifications

Phosphorylation on Tyr-10 modulates pumping activity.
  • Cellular localization

Cell membrane. Melanosome. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.
  • Target information above from: UniProt accession P05023 The UniProt Consortium

IHC image of Ab7671 staining in Human Kidney Carcinoma formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with Ab7671, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Question

Thank you very much for the reply.
I'm working with membrane samples, would they also be heated to 95-100C for 10 minutes?
How come the datasheet for this product warns "Do not boil the protein samples, as boiling causes aggregation of the Na,K-ATPase
(60 degrees Celcius appears to work fine). The aggregate band will appear at ˜150 kDa on Western Blots." if samples can be boiled?
Kind regards,

Answer

Thank you for your response.

Our Lab regularly test this product on whole cell lysates and heat the samples 95-100C for 10 minutes. The signal is very clear and distinctive. My personal experience is that plasmamebrane protein can be heating for 5 min. As I have mentioned in my previous e-mail, this comment was published long time ago part of a customer's feedback.

If you need any further assistance, please do not hesitate to contact me.
