    Alpha-2 adrenergic receptors mediate the catecholamine-induced inhibition of adenylate cyclase through the action of G proteins. The rank order of potency for agonists of this receptor is oxymetazoline > clonidine > epinephrine > norepinephrine > phenylephrine > dopamine > p-synephrine > p-tyramine > serotonin = p-octopamine. For antagonists, the rank order is yohimbine > phentolamine = mianserine > chlorpromazine = spiperone = prazosin > propanolol > alprenolol = pindolol.

  • ab45871 staining alpha 2a Adrenergic Receptor in mouse DRG neurons by Immunocytochemistry/ Immunofluorescence. The ganglia were collected from postnatal day 0–3 pups of C57 BL/6 mice and enzymatically dissociated with medium containing 0.25% trypsin at 37°C. Dissociated neurons were then plated onto glasscoverslips and maintained at 37°C in culture media. After drug treatment, the cultures were fixed with 4% paraformaldehyde for 10 min, washed in phosphate-buffered saline (PBS), permeabilized, and preblocked with PBS containing 10% normal donkey serum and 0.1% Triton X-100 for 2 h at room temperature. The cultures were then incubated overnight at 4 °C with the primary antibodies. A Biotin-conjugated anti-goat IgG and Cy3-conjugated streptavidin was used to detect alpha 2a adrenergic receptor.
