Thank you for taking time to complete our questionnaire. I am sorry to hear that this antibody is not providing satisfactory results. The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality. Having reviewed this case, I would like to offer some suggestions to help optimize the results from ab21607. I would also appreciate if you can confirm some further details: In many cases, antigenicity of elastic fiber components can be enhanced by treating with 6 M guanidine HCl, 50 mM dithiothreitol, 20 mM Tris pH 8.0 for 15 minutes. After washing with 20 mM Tris pH 8.0, the slides are treated with 100 mM iodoacetamide in the dark for 15 minutes, washed and stained using normal protocols. I would try including this step. A number of other researchers on IHC world have stained Elastin successfully be using proteinase K antigen retrieval at 37°C for 20 min. This is another step I would try but without the pretreatment detailed above. How was the antibody treated prior to storage at -20°C? Glycerol should be added to a final volume of 50% prior to storage. Should the suggestions not improve the results, please do let me know.
