Application Abreviews Notes
ELISA 1/10000 - 1/50000.
WB 1/1000 - 1/5000. Predicted molecular weight: 10 kDa.

  • Function

Selectively inhibits insulin-stimulated glucose utilization and glycogen deposition in muscle, while not affecting adipocyte glucose metabolism.
  • Sequence similarities

    Belongs to the calcitonin family.
  • Domain

The mature protein is largely unstructured in the absence of a cognate ligand, and has a strong tendency to form fibrillar aggregates. Homodimerization may be the first step of amyloid formation.
  • Post-translational

Amyloid fibrils are degraded by IDE.
  • Cellular localization

Customer reviews and Q&As

1-2 of 2 Abreviews or Q&A

Thank you for your enquiry and your interest in our products.
Concerning ab18018 - there is significant sequence homology between human amylin and that of mouse and rat, but this antibody has only been tested with human samples.
Unfortunately, we do not have any experimental data regarding the cross-reactivity of ab15125 and ab15128 and ab55411 with mouse or rat samples too.
I compared the immunogen for ab15125 with that of the mouse and rat Amylin proteins and the percentage homology for both is 100%. Based on that information ab15125 is most likely not suitable for your application.
Please do not hesitate to contact me with additional questions or concerns.

1)Antibody code ab18018 2)Batch number 96335 3)Purchase order number or preferably Abcam order number our purchase order number was 7150 but I think you may put six zeros before that. It was ordered via the Babraham Institute stores. 4)General Information Antibody storage conditions (temperature/reconstitution etc) Arrived at ambient temperature, stored at 4oC overnight, used the following day and the day after. 5)Description of the problem (high background, no signal, non-specific color development, poor standard curve etc.) No signal or weak signal Type of ELISA (Direct ELISA/Indirect ELISA/Sandwich ELISA etc.) Indirect 6)Sample (Species/Cell type/Cell line etc.) Synthetic peptides 7)Coating well (Buffer/Concentration of the coating material etc.) Bicarbonate buffer. Peptide concentration 0.1ng-50ug/well coated O/N 4oC 8)Blocking conditions (Buffer/time period, Blocking agent etc.) 5% Milk/PBS/0.05%Tween 9)Primary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step) Ab18018 (the antibody in question), in blocking buffer above at 1:1000-1:100000, 1hr RT Washed in PBS/0.05% Tween x5 10)Secondary Antibody (Manufacturer/Species/Diluent/Dilution/Incubation time, Wash step) Goat anti-rabbit IgG whole molecule HRP Sigma in blocking buffer Wash as above x5 11)Detection method (Substrate/Diluent etc.) TMB 12)Positive and negative controls used (please specify) Mouse R10/99 mAb and our own polyclonal rabbit anti-human Ab both work extremely well. 13)Optimization attempts (problem solving) How many times have you tried the ELISA? Twice 14)Have you run a "No Primary" control? Yes 15)Do you obtain the same results every time? Yes. 16)What steps have you altered? Concentration of coating material, peptide species and length, concentration of the antibody, 17)Additional Notes Two experiments were carried out by different scientists on different days. Used different batches of various peptides for coating. Please don't hesitate to contact me again if you require more information.

Thank you very much for answering our questions. We would like to recommend the following modifications to your protocol: incubate the primary antibody overnight at 4C and try a 1-3% BSA block instead of milk. As the antibody is polyclonal it will recognise a variety of sites on the protein and this maybe less specific than ab11022 and hence require longer incubation times. You mention "synthetic peptides" as your samples, are those human sequences (the antibody has only been tested against human amylin)? We would also suggest doing a dot blot to check that the antibody binds to the peptides. Please do not hesitate to contact us again if you need further advice and be assured that should you still have problems in the recommended conditions we will offer you a replacement vial.
