By Western blot, this antibody detects an ~50 kDa protein representing FIAF from rat adipose tissue extracts.

  • Function
    Protein with hypoxia-induced expression in endothelial cells. May act as a regulator of angiogenesis and modulate tumorgenesis. Inhibits proliferation, migration, and tubule formation of endothelial cells and reduces vascular leakage. May exert a protective function on endothelial cells through an endocrine action. It is directly involved in regulating glucose homeostasis, lipid metabolism, and insulin sensitivity. In response to hypoxia, the unprocessed form of the protein accumulates in the subendothelial extracellular matrix (ECM). The matrix-associated and immobilized unprocessed form limits the formation of actin stress fibers and focal contacts in the adhering endothelial cells and inhibits their adhesion. It also decreases motility of endothelial cells and inhibits the sprouting and tube formation.

Thank you fro bringing this to our attention. Are you seeing a band at 50 kDa, and if so, is it more intense than the other bands? Where do the other bands appear in the blot? If you can attach an image, the may help me understand what is going on.

It is possible the antibody is defective but we ask our customer to provide a few details of their protocol, in the hope that we can suggest a modification that will improve the result. If not, I will send a refund.

Can you please tell me how the samples are prepared, in particular, whether or not you are using protease inhibitors? Are the samples reduced and denatured?

How is the membrane blocked before incubation with the antibody? And finally, are you confident that the secondary antibody is effective and not reacting with proteins in the sample?

We do not assign strict expiration dates for our products, however we follow general guidelines: Antibodies stored at 4C may be viable up to 1 year, those stored at -20C may be viable for up to 5 years, and those stored at -80C may be viable for up to 10 years. Thank you for your enquiry. The predicted KDa that this target should run at (Swiss Prot) is 46KDa, there is no indication of an alternate isoform of this target that I have been able to source. The paper cited below appears to indicate that the full length target observed usually as a single band at 45KDa is the unprocessed form; however proteolytic cleavage is also reported in this paper. Where this occurs the author suggests that a band of 25-30KDa may be observed. http://www.jbc.org/cgi/reprint/M403058200v1.pdf I am unsure as to the conditions that dictate the cleavage for this target; however it does sound like you may be detecting both forms. Since the rabbit is a pab (CQGPKGKDAPFKDSE) and will detect multiple epitopes it is possible that both forms may be detected. Thank you for your enquiry and patience. To our knowledge, this antibody has only been tested for application in Western blotting, and so we don't have any information about how ab2920 works in IP. Have you done a positive control? I would suggest running a Western with rat adipose tissue extract to ensure that the antibody is functioning properly. Also, when did you purchase this antibody? What is the lot number (located on the vial)? On the datasheet it is recommended to store this antibody at +4°C short term and aliquot and store at -20°C or -80° for longer term storage.
