    Defects in AGTR1 are a cause of renal tubular dysgenesis (RTD) [MIM:267430].   • ab18801 staining Angiotensin II Type 1 in porcine kidney tissue by Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections).

    Tissue was fixed with paraformaldehyde and blocked with 5% BSA for 30 minutes at 20°C; antigen retrieval was by heat mediation with a citrate buffer. Samples were incubated with primary antibody (1/100 in PBS + Triton X-100 + goat serum) for 24 hours at 4°C. An HRP secondary antibody was used.

Este anticuerpo esta testado y garantizado para reconocer el receptor Angiotensin II Type 1 mediante WB, IHC-Fr, IHC-FoFr, IHC-P y microscopia electrónica en ratón, rata, bovino, canino y cerdo.

Es necesario sin embargo tener en cuenta el nivel de expresión del receptor en los distintos tejidos, ya que es posible que el motivo por el que este anticuerpo y los de otras marcas no dieron tinción en los tejidos mencionados.

Por ello siempre es necesario teñir en paralelo con las muestras un control positivo en el que se tenga la certeza de la expresión del antígeno, y poder así comparar con el resultado en la muestra.

Respecto a este receptor, atendiendo a su perfil de expresión, es cierto que no está presente en los tejidos indicados (hígado, riñón y corazón). En cambio, en tejidos de cerebro, piel y tejido blando el perfil de expresión es mayor.

Mi recomendación es que si el cliente tiene la posibilidad, use estos tejidos como control positivo, y si siguiera sin observar tinción ninguna nos contacte de nuevo para reemplazarle o reembolsarle el producto.

We did not receive any recent complaints for these lot so I am sorry we are unable to determine the cause of the problem. GR64320-2 is most recent batch for this product which was also failed along with GR13003-3 so I believe that the problem could be with protocol rather than antibodies. I would suggest trying the troubleshooting as recommended below even if the previous lot worked.

- Try the antigen retrieval step. 5 minutes microwave with citrate buffer pH 6.0 could help.
- Consider including 0.3 M glycine in the blocking buffer, before applying the primary antibody. Glycine will bind fre aldehyde groups that would otherwise bind the primary and secondary antibodies, leading to high background. Background due to free aldehyde groups is more likely to occur when the fixative is glutaraldehyde or paraformaldehyde.
- Wash the sections with PBST rather PBS
- Use 10% NDS for blocking
- Try no primary control in order to check the non specific staining due to secondary antibody.

The protcol looks fine to me however could you please clarify the following;

- Is it true no antigen retrieval was used?
- Are the tissue sections frozen or paraffin embedded? If they were frozen then acetone fixation is recomended. If they are PFA fixed and paraffin embedded then antigen retrieval step is recomended. Could you summarize the protocol steps.
- Does the antibopdy unable to stain the target or it was showing high background e.g. non specific binding.
- Could you provide images if possible?

Immunohistochemistry results will vary with different tissue types and fixation protocols. We recommend testing a range of three dilutions, to determine which gives the most intense specific signal with the least background staining. I suggest 1/50, 1/150, and 1/450. If you have to skip this test, then I suggest using 1/200 with the Vectastain kit.
