Thank you for having your post-doc provide the protocol and image. Is she confident that the samples are not degraded? The smear around the 49kDa marker suggest that might be the case, but I do not have an explanation for absence of signal for the recombinant protein or the HepG2 samples, unless the recombinant protein is a fragment that does not include the immunogen sequence that the antibody was generated against. She mentioned that a different antibody does detect this recombinant protein. Was that antibody also tested against the serum and HepG2 lysates?

I think the protocol is correct, so I think either the samples are the problem, or the antibody. I expect the post-doc has tested the same samples with other antibodies that give good results. If she is confident that the samples are good, then I suggest we send a replacement, a different antibody against the same protein. I recommend ab126718.

We are not aware of any problems with this antibody. We have also not received any negative feedback in this regards so far.
Indeed, we have not yet tested this antibody on the mouse material. Even though through sequence homology we predict the antibody to work on mouse material, this would need to be assessed experimentally. Do you have any possibility to test this antibody on human material as a positive control?
I do however appreciate the time you have spent in the laboratory and understand your concerns. It is regrettable the results have not been successful so far. I agree that with your protocol and with the experience in your laboratory the results should have been positive. Therefore it either is that the antibody does not crossreact with mouse or has been damaged. Please do let me know whether you have the possibility to test this antibody on human material.
