Involvement in diseaseGenetic variations in TNFSF4 influence susceptibility to systemic lupus erythematosus (SLE) [MIM:152700]. SLE is a chronic, inflammatory and often febrile multisystemic disorder of connective tissue. It affects principally the skin, joints, kidneys and serosal membranes. It is thought to represent a failure of the regulatory mechanisms of the autoimmune system. Note=The upstream region of TNFSF4 contains a single risk haplotype for SLE, which is correlated with increased expression of both cell-surface TNFSF4 and TNFSF4 transcripts. Increased levels of TNFSF4 are thought to augment T cell-APC interaction and the functional consequences of T cell activation, thereby destabilizing peripheral tolerance.

Flow cytometry overlay histogram showing positive HUVEC cells (left panel) and negative HeLa cells (right panel) stained with ab267561 (red line). The cells were incubated in 1x PBS containing 10% normal goat serum to block non-specific protein-protein interaction followed by the antibody (ab267561) (1x 106 in 100 µl at 5 µg/ml (1/100)) for 30 mins on ice. Isotype control antibody (black line) was Rabbit IgG (monoclonal) Allophycocyanin (ab232814) used at the same concentration and conditions as the primary antibody. Unlabelled sample (blue line) was also used as a control.
Acquisition of >5000 events were collected using a 40 mW Red laser (640nm) and 670/14 bandpass filter.
