  • Product overview

Apoptotic DNA Ladder Isolation Kit (ab65627) provides an easy and sensitive solution for detecting DNA fragmentation in apoptotic cells and tissues. The new procedure selectively extracts DNA ladders without interference of intact genomic DNA in only 2-4 hours. As little as 5% of apoptotic cells in a sample can be visualized with this high sensitivity assay.

Properties

Images

  • Apoptosis induced in Jurkat cells (1e6 cell x mL-1) by incubation for 24 hours with 1) Control medium; 2) 2 uM Camptothecin (ab120115); 3) 10 uM Camptothecin; 4) 10 uM Camptothecin + 20 nM caspase inhibitor Z-VAD(OMe)-FMK (zVAD; ab120487); 5) 50 ng x mL-1 anti-Fas Ab (MBL), 6) 0.05 mM H2O2 (Sigma); 7) 1 mM H2O2. 5 uL of DNA preparation loaded into each well.

  • DNA ladders were extracted from 2 x 106 Jurkat cells were treated with 2 μM Camptothecin. The kit extracts laddered DNA only but not intact genomic DNA; therefore, more cells can be extracted and load on the agarose gel. Lane 1: 1 kb DNA ladder; Lane 2: 6 hour induction; Lane 3: 12 hour induction.

Answer

Thank you for contacting us. Please find below the reply from the lab.

The DNA suspension buffer already contains glycerol to help keep the sample in the well. The electric current should not be started immediately after adding the sample in the well but you should wait for a few minutes to allow the sample to settle down in the well. We have never experienced this after using 30ul of the suspension buffer to resuspend the DNA pellet. You could add some more suspension buffer, or simply add 1-5ul glycerol to the sample in suspension buffer to avoid floating out of the well.

Answer

Thank you very much for your email. All Abcam products are thawed to room temperature before shipping and all are shipped out at room temperature every day of the week. Please note that the majority of enzymes are stable at room temperature (RT) for long periods of time. Experiments conducted at Abcam show that, for enzymes left at RT, activity remains constant for a long time before showing any decline. However, as a precautionary measure and to extend the shelf life of the products, we ship our products in refrigerated packaging which has been shown to provide a stable environment for antibodies in transit. As part of our Abpromise, our policy is that if a product does not work as specified on the datasheet, we will offer a replacement or reimbursement if you contact us within 120 days of purchase. Once you have received the vial please store it as per the instructions on the datasheet. I hope this information helps but please don't hesitate to contact me for additional information or assistance.
