    Forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.

  • Immunohistochemical staining of aquaporin 1 in rat brain.
  • Overlay histogram showing HEK293 cells stained with ab9566 (red line). The cells were fixed with methanol (5 min) and then permeabilized with 0.1% PBS-Triton for 20 min. The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions followed by the antibody (ab9566, 2µg/1x106 cells) for 30 min at 22°C. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22°C. Isotype control antibody (black line) was mouse IgG2b [PLPV219] (ab91366, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in HEK293 cells fixed with 4% paraformaldehyde (10 min)/permeabilized in 0.1% PBS-Triton used under the same conditions.

The Immunogen for ab9566Anti-Aquaporin 1 antibody [1/22] is provided:

GQVEEYDLDDDINSRVEMKPK

This has 9% alignment to mouse Aquaporin 2. I am sorry to confirm that the antibody has not specifically been tested for crossreactivity to aquaporin 2 in the laboratories. However, the alignment to aquaporin 1is very low and we would not expect this antibody to cross react with aquaporin 2

For ab15116 Anti-Aquaporin 2 antibody,the immunogen for this antibody is a peptide comprised of a 15 amino acid sequence from near the C-terminus of aquaporin 2. The 15 amino acid are within the last 50 amino acids of the sequence (NP_037041). Again, I am sorry this antibody has not been specificallytestedfor crossreactivity to aquaporin 1. However, there is again very low alignment to the immunogen to aquaporin 1 so we would not expect it todetect aquaporin 1.

Thank you for your enquiry. I contacted the authors who made the antibody and Prof. Peter Nemeth told me, "We used the DDD sequence by technical difficulties of peptide synthesis. The one amino acid difference in antigen didn’t influence the specificity of our antibody according to the results of different testing techniques."
