  • Function

    Forms a water-specific channel that provides the plasma membranes of red cells and kidney proximal tubules with high permeability to water, thereby permitting water to move in the direction of an osmotic gradient.

    Expressed in a number of tissues including erythrocytes, renal tubules, retinal pigment epithelium, heart, lung, skeletal muscle, kidney and pancreas. Weakly expressed in brain, placenta and liver.

Answer

I'm sorry to hear you are experiencing problems with ab11025. This antibody has not been tested in immunofluorescence and this may be the reason for the problem. I would recommend trying a biotin-ABC-DAB detection system which is more sensitive that IF (indeed I would recommend more powerful fluorochromes in general, such as Alexa or Cy3 antibodies for immunofluorescence detection). I would also recommend the following modifications: -incubate the antibody for longer, such as overnight at 4C -use tritonx100 in the dilution buffer for the blocking agent and the primary and secondary antibodies -you did not mention the fixation time or temperature, can I please confirm that the alcohol should be ice cold (pre-cooled at -20C then incubation should be at RT) and the fixation time should be around 10min.
