|WB||Use a concentration of 1 µg/ml. Detects a band of approximately 48 kDa (predicted molecular weight: 35 kDa).|
FunctionForms a water-specific channel. Osmoreceptor which regulates body water balance and mediates water flow within the central nervous system.
Tissue specificityBrain - muscle >> heart, kidney, lung, and trachea.
Sequence similaritiesBelongs to the MIP/aquaporin (TC 1.A.8) family.
DomainAquaporins contain two tandem repeats each containing three membrane-spanning domains and a pore-forming loop with the signature motif Asn-Pro-Ala (NPA).
modificationsPhosphorylation by PKC at Ser-180 reduces conductance by 50%. Phosphorylation by PKG at Ser-111 in response to glutamats increases conductance by 40%.

Predicted band size: 35 kDa
Observed band size: 48 kDa why is the actual band size different from the predicted?
Additional bands at: 125 kDa (possible non-specific binding), 90 kDa (possible non-specific binding)
Exposure time: 30 seconds
The predicted molecular weight of Aquaporin 4 is 34 kDa (SwissProt), however we expect to observe a banding pattern at 48 kDa. Abcam welcomes customer feedback and would appreciate any comments regarding this product and the data presented above.
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab156924 overnight at 4°C. Antibody binding was detected using an anti-rabbit antibody conjugated to HRP, and visualised using ECL development solution ab133406
