Question

We have purchased some antibodies against rat platelets:

Primary:

ab108949 - mouse monoclonal anti CD63, FITC labeled
(ab106163 as isotype control)

ab54427 - mouse monoclonal anti CD62P
(ab91353 as the isotype control, as a secondary antibody)

ab95780 anti CD42d armenian hamster antibody
(ab18479 as an isotype control, for which we are using APC-conjugated Goat Anti-Hamster IgG from Leinco).

We are ibodies.doing flow cytometry experiments on fixed whole blood, and fixed platelet-rich plasma. The problem is that the samples with the isotype control show the same fluorescence as the samples with the primary ant We have tried a number of different fixing and staining protocols, with and without washing, but until now had no success. What I am wondering about is, how reliable are these primary antibodies for the use with rat platelets?

Some help would be greatly appreciated.

Answer

Thank you for your reply.

I am sorry that you have been having issues with some of our antibodies in Flow Cytometry. Under our Abpromise we guarantee that ab108949, ab54427 & ab95780 should work in flow cytometry on rat samples. If we cannot resolve the issue you are having with the antibody then I would be happy to either send a replacement antibody or to process a refund.

To be able to help you further in this matter, would you be able to send me a copy of the protocol that you have been using, along with any modifications that you have tried, to see if there is anything I can suggest.

I look forward to your reply and helping resolve this matter.

Question

I selected the following antibodies from the ABcam catalog:

Primary:

Anti-CD63 antibody [AD1] (FITC)

ab108949

$337.00

Anti-CD62P antibody [Psel.KO.2.7]

ab54427

$337.00

Anti-CD42d antibody [1C2]

ab95780

$337.00

Secondary:

Goat anti-Mouse IgG H&L (Cy5.5 ®)

ab6947

$185.00

Goat anti-Mouse IgG1 heavy chain (FITC)

ab97239

$165.00

Goat polyclonal to Armenian Hamster (TR)

ab5743

$165.00

Isotype controls:

MOUSE IgG1 antibody [MOPC-21] (FITC)

ab106163

$325.00

Mouse IgG1 [ICIGG1] - Isotype Control

ab91353

$315.00

Armenian Hamster IgG monoclonal [HTK888] - Isotype control

ab18479

$325.00

Two questions:

For the secondary antibody, will the first one (ab6947) work against ab54427, or do we need to buy the second one (ab97239)? One is against IgG, the other one is against IgG1. ab54427 is IgG1.

If you could double-check the selection for compatibility (e.g., correct isotype controls) and then ask the sales department to offer us a quote, that would be great.

The antibodies you selected look good to me. To generate a quote we do need to have a shipping address and in your last email, you mentioned that you were at the University of Toronto at the moment. Could you please send me your current email address and I will generate the quote.

I look forward to your reply.

Answer

Thank you for contacting us with your question. I contacted the production lab to enquire about the isotype of ab36436, and unfortunately they said that the antibody has not been characterized to distinguish if it is IgG1 or IgG2.   We do have hamster IgG isotype controls that you may use as control antibodies. I would recommend ab18479, which is an Armenian Hamster IgG isotype control. This antibody has been tested on a variety of mouse, rat, and human tissues. I hope this information is helpful, but please let me know if you have further questions.
