Properties

Applications

Our Abpromise guarantee covers the use of ab49355 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
WB Use a concentration of 0.6 µg/ml. Detects a band of approximately 46 kDa (predicted molecular weight: 33 kDa). Good results were obtained when blocked with 5% non-fat dry milk in 0.05% PBS-T.
IHC-P Use a concentration of 5 µg/ml. Perform heat mediated antigen retrieval with citrate buffer pH 6 before commencing with IHC staining protocol.

Target

  • Function

Mediates the endocytosis of plasma glycoproteins to which the terminal sialic acid residue on their complex carbohydrate moieties has been removed. The receptor recognizes terminal galactose and N-acetylgalactosamine units. After ligand binding to the receptor, the resulting complex is internalized and transported to a sorting organelle, where receptor and ligand are disassociated. The receptor then returns to the cell membrane surface.
  • Tissue specificity

Expressed exclusively in hepatic parenchymal cells.
  • Sequence similarities

    Contains 1 C-type lectin domain.
  • Post-translational
    modifications

Phosphorylated on a cytoplasmic Ser residue.
  • Cellular localization

    Membrane.

    Secondary
HRP conjugated anti-Rabbit IgG at 1/50000 dilution

    Predicted band size: 33 kDa
    Observed band size: 46 kDa
why is the actual band size different from the predicted?

  • IHC image of ab49355 staining in human normal liver formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol F. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab49355, 5µg/ml, for 15 mins at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.
