My question is, at what time point do I read my standard curve and generate the standard curve equation. At 10 min? At 60? Or does it not matter. I ask because this assay is more of a kinetic assay and it should vary over time.

It is recommended to measure standard curve at both times 10 mins and 60 mins. Then use the difference in Abs at 60min - Abs at 10min to plot the Std curve (OD on Y axis versus Glutamate nmol on the X axis).
Please let me know if you have any further questions.
