Target

  • Function

Catalytic component of a P4-ATPase flippase complex which catalyzes the hydrolysis of ATP coupled to the transport of aminophospholipids from the outer to the inner leaflet of various membranes and ensures the maintenance of asymmetric distribution of phospholipids. Phospholipid translocation seems also to be implicated in vesicle formation and in uptake of lipid signaling molecules (Probable). May be involved in the uptake of farnesyltransferase inhibitor drugs, such as lonafarnib.
  • Sequence similarities

Belongs to the cation transport ATPase (P-type) (TC 3.A.3) family. Type IV subfamily.
  • Cellular localization

Cell membrane. Early endosome. Recycling endosome. Endoplasmic reticulum. Exit from the endoplasmic reticulum requires the presence of TMEM30A, but not TMEM30B. In the presence of TMEM30A, predominantly located in the plasma membrane.

    Answer

Thank you for your reply and thank you for trying the mouse samples as well! I am very sorry that these3 antibodies did not perform as expected. I would be happy to offer you either a free of charge replacement for each antibody, or a credit or refund. For AT11A, we only have on other antibody in the catalog (ab117577) which has not yet been tested in mouse. For AT11B, we 2 other antibodies that are predicted to work in mouse, but also have not been tested with this species: ab82640 and ab117580. For AT11C, we have no other antibody available. I can offer you a credit or refund in this case or a replacement with an antibody to a different protein. In case of AT11A and AT11B, we are not able to guarantee the replacement antibodies for mouse (as they have not been tested), but I can give you a testing discount code for each replacement antibody. Alternatively, a different lot can be tried for ab105377 (AT11B). For the other 2 antibodies we currently have no vial in stock and I am not sure if the same or a new lot would come in next. Some more information on our testing discount program, should you decide to chose the antibodies that are not yet tested in mouse: For UNTESTED species and/or applications, we have established a testing discount program. Here is a brief description of how it works: The testing discount program is for customers who like to use an antibody/protein on an untested species/application. You would purchase the antibody at full price, test it and submit an Abreview with your data (positive or negative). On your next order you will receive a discount for ONE antibody at the full price (100%) of the antibody you have tested. The terms and conditions applicable to this offer can be found here: www.abcam.com/collaborationdiscount. Please let me know how you would like to proceed and I'd be happy to arrange it for you. I look forward to hear back from you and resolve this case for you.

    Answer

Thank you for your phone call. I am sorry to hear that these antibodies are not providing satisfactory results so far. The details provided will enable us to investigate this case and will provide us with vital information for monitoring product quality. Having reviewed this case, I would like to offer some suggestions to help optimize the results, as we discussed over the phone: 1) Since bovine samples have not been tested we cannot guarantee the antibodies for this species. However, mouse samples have been tested and you said you have mouse samples available to you. Thus, trying the antibodies on the mouse samples would be advisable to do. 2) Since high background was observed, using 5% milk for 2h (instead of 1% milk for 30 min) could help improve the signal and reduce the background. 3) Also, diluting the primary antibody in milk blocking solution can help as well. Should these suggestions not improve the results, please do let me know. In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund. I hope this information is helpful, and I thank you for your cooperation.
