Application Abreviews Notes
ICC/IF Use at an assay dependent concentration. Used at a dilution of 1/1000 for 1 hr on HeLa cells (see Abreview).
IHC-P 1/250 - 1/1000.
WB 1/4000. Detects a band of approximately 48 kDa (predicted molecular weight: 48 kDa). Highly expressed in testis and weakly in skeletal muscle, thymus and spleen. Also highly expressed in colon, ovarian, prostate, neuroblastoma, breast, pancreas and cervical cancer cell lines. Expression is cell-cycle regulated, low in G1/S, accumulates during G2/M, and decreases rapidly after.
IP Use a concentration of 2 - 10 µg/ml.


  • Function

    Contributes to the regulation of cell cycle progression. Required for normal mitosis. Associates with the centrosome and the spindle microtubules during mitosis and functions in centrosome maturation, spindle assembly, maintenance of spindle bipolarity, centrosome separation and mitotic checkpoint control. Phosphorylates numerous target proteins, including ARHGEF2, BRCA1, KIF2A, NDEL1, PARD3, PLK1 and BORA. Regulates KIF2A tubulin depolymerase activity (By similarity). Required for normal axon formation. Plays a role in microtubule remodeling during neurite extension. Important for microtubule formation and/or stabilization.

    Highly expressed in testis and weakly in skeletal muscle, thymus and spleen. Also highly expressed in colon, ovarian, prostate, neuroblastoma, breast and cervical cancer cell lines.
  • Sequence similarities

    Belongs to the protein kinase superfamily. Ser/Thr protein kinase family. Aurora subfamily.
    Contains 1 protein kinase domain.
  • Post-translational

    Activated by phosphorylation at Thr-288; this brings about a change in the conformation of the activation segment. Phosphorylation at Thr-288 varies during the cell cycle and is highest during M phase. Autophosphorylated at Thr-288 upon TPX2 binding. Phosphorylated upon DNA damage, probably by ATM or ATR.
    Ubiquitinated by CHFR, leading to its degradation by the proteasome (By similarity). Ubiquitinated by the anaphase-promoting complex (APC), leading to its degradation by the proteasome.
  • Cellular localization

    Cytoplasm > cytoskeleton > centrosome. Cytoplasm > cytoskeleton > spindle pole. Detected at the neurite hillock in developing neurons (By similarity). Localizes on centrosomes in interphase cells and at each spindle pole in mitosis.

    35S-Met labeled whole cell lysate from cells transfected with a human Aurora A expression construct. ab1287 was used at 4 µg/ml for IP. Detection: Autoradiography.
  • ab1287 (1/250) detecting Aurora A in paraffin embedded Human pancreatic tumour tissue sections. Detection: DAB staining;
  • ab1287 at a 1/1000 dilution staining synchronised HeLa cells by ICC/IF. The cells were paraformaldehyde fixed and blocked with 1% serum prior to incubation with the primary antibody for 1 hour. A Cy5 conjugated donkey anti-rabbit antibody was used as the secondary. The image shows a DAPI panel, Aurora-A panel, and merge panel of DAPI (blue), microtubules (red) and Aurora-A (green).

In general, we recommend the tissue you see on the datasheet, in this case a pancreatic tumor. We offer sections of pancreatic adenocarcinoma, not of the exact same tumor, but we assume the staining will be similar. Here is a link to the datasheet for the sections.

For ab2254, we would suggest microwave pre-treatment in citrate buffer (pH 6.0) for 5 mins high then 5 mins simmer (800W conventional microwave). We would suggest blocking the non-specific binding sites with 1%BSA for 1 hr and incubating the samples with the primary antibody for 2 hrs (40 min is too short). This antibody ahs been tested for IHC using human pancreatic tumor sections, so it may be worth using this type of tissue sections as positive control.
