  • Function
    Contributes to the regulation of cell cycle progression. Required for normal mitosis. Associates with the centrosome and the spindle microtubules during mitosis and functions in centrosome maturation, spindle assembly, maintenance of spindle bipolarity, centrosome separation and mitotic checkpoint control. Phosphorylates numerous target proteins, including ARHGEF2, BRCA1, KIF2A, NDEL1, PARD3, PLK1 and BORA. Regulates KIF2A tubulin depolymerase activity (By similarity). Required for normal axon formation. Plays a role in microtubule remodeling during neurite extension. Important for microtubule formation and/or stabilization.

    Highly expressed in testis and weakly in skeletal muscle, thymus and spleen. Also highly expressed in colon, ovarian, prostate, neuroblastoma, breast and cervical cancer cell lines.

    Activated by phosphorylation at Thr-288; this brings about a change in the conformation of the activation segment.     Ubiquitinated by CHFR, leading to its degradation by the proteasome (By similarity). Ubiquitinated by the anaphase-promoting complex (APC), leading to its degradation by the proteasome.

I'm sorry to hear you are having a problem with ab3849. I realize that you do not have problems with other antibodies, however, as I mentioned for ab7031, the conditions for each antibody can be different and I believe with some suggestions the antibody will work well for you. I would like to suggest the following modifications to your protocol: because you are seeing too many bands, 1) the recommended dilution for this antibody is actually 1/5000 to 1/20,000 as stated on the datasheet. For the blot on the datasheet the dilution was 1/12,000. I would try a dilution in this range, perhaps 1/20,000. 2) Incubate in primary antibody for 90 minutes at room temperature with 0.5% to 5.0% milk, 3) Incubate in secondary antibody for 60 minutes at room temperature with 0.5% to 5.0% milk, 4) block in 5% milk overnight at 4C.
