  • ab121794, at 1/50 dilution, staining B7-H6 in paraffin-embedded Human gallbladder tissue, by Immunohistochemistry.
  • ab121794 at 1/50 dilution staining a cell-clot mixture of JVM (B7-H6 neg) and Hela (B7-H6 pos) (5:1) cells by IHC-P. The cell matrix section was subjected to heat-mediated antigen retrieval using 10 mM Citrate Buffer pH6 before being blocked with 10% horse serum for 30 minutes at 23°C and then being incubated with ab121794 (1/50 dilution) for 16 hours at 4°C. The secondary used was a Biotin conjugated goat anti-rabbit polyclonal (1/200 dilution).

I have discussion with our IT department - unfortunately they do not save the previous versions of datasheets when updating them. The online datasheets along with pdf's gets updated every 15-20 days which is necessary for consistency between the two. I am sorry we will unable to provide you the version older than 5th Nov 2012.

The pdf's gets updated irrespective of changes the datasheet team makes for example the new version will be exactly the same if no information was added or deleted.

Please note that we do track the data we change on the datasheet so if there is any difference between the old and new, we do know what has been changed. For ab121794 WB application was deleted as we discussed on phone.

As far as we are aware, this antibody has also not been added into any of the news letter.

Whenthe laboratory characterized thisantibody, there was almost no literature available to compare the staining results with, but now there is some new literature describing the localization of the target protein. The laboratory has decided to re-evaluate this antibody andwe willget back to you whenwe hear the results from the this evaluation.



Attached is also an image we had in colorectal cancer tissue.



Here just FYI - the original test were based on theannotation summary for Normal Human Tissues :

"A majority of normal tissues showed weak to moderate cytoplasmic and nuclear positivity. Strong staining was observed in glandular cells in adrenal cortex and gall bladder. Most cell in CNS and spleen were negative." And for Human Cancer Tissues:
"A majority of colorectal and urothelial cancers showed moderate to strong cytoplasmic and nuclear positivity while a majority of prostate and several breast cancers displayed moderate cytoplasmic staining. Remaining malignancies were weakly stained or negative."

Bisher wurde der ab121794 Anti-B7-H6 antibody leider nicht in ELISA getestet, und auch eine mögliche Kreuzreaktionen mit anderen B7-H Molekülen wurde nicht experimentell untersucht.

Der Antikörper wurde in WB getestet, aber es konnte leider kein Signal detektiert werden:

"The Western blot analysis for each antibody is performed using an identical set up with no optimization, i.e. the same five protein lysates, as well as the same gel type and same conditions are used for all antibodies. As the protein samples and running conditions are not specifically selected and adapted for each antibody and its corresponding protein, not every antibody has been successfully validated for the Western blot application."

Da der Antikörper sein Zielprotein in der IHC erkannte, kann dies ein Indiz dafür sein, dass der Antikörper nur mit dem nativen, nicht-reduzierten Protein reagiert.
