Application Abreviews Notes
WB 1/1000. Detects a band of approximately 80 kDa (predicted molecular weight: 62 kDa).
ICC/IF 1/60.
Flow Cyt 1/60.
  • Application notes
Is unsuitable for IHC-P or IP.
  • Target

    • Function

Inhibits the chaperone activity of HSP70/HSC70 by promoting substrate release. Has anti-apoptotic activity.
    • Involvement in disease

Defects in BAG3 are the cause of myopathy myofibrillar BAG3-related (MFM-BAG3) [MIM:612954]. A neuromuscular disorder that results in early-onset, severe, progressive, diffuse muscle weakness associated with cardiomyopathy, severe respiratory insufficiency during adolescence, and a rigid spine in some patients. At ultrastructural level, muscle fibers display structural alterations consisting of replacement of the normal myofibrillar markings by small, dense granules, or larger hyaline masses, or amorphous material.
    • Sequence similarities

      Contains 1 BAG domain.
      Contains 2 WW domains.

    • Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HeLa (human cervix adenocarcinoma epithelial cell) cells labeling Bag3 with ab209251 at 1/60 dilution, followed by Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) secondary antibody at 1/1000 dilution (green). Confocal image showing cytoplasmic and nuclear staining in HeLa cell line. The nuclear counter stain is DAPI (blue). Counterstained with ab195889 Anti-alpha Tubulin antibody [DM1A] - Microtubule Marker (Alexa Fluor® 594) at a 1/200 dilution (red). The control is the secondary antibody only.

Negative control cell line: HL-60 (PMID: 17974966).

    • All lanes : Anti-Bag3 antibody [EPR20207-109-3] (ab209251) at 1/1000 dilution

Lane 1 : Human skeletal muscle lysate
Lane 2 : HeLa (human epithelial cell line from cervix adenocarcinoma) whole cell lysate
Lane 3 : Mouse heart lysate
Lane 4 : Rat heart lysate
Lane 5 : HL-60 (human acute promyelocytic leukemia promyeloblast) whole cell lysate

Lysates/proteins at 20 µg per lane.

      All lanes : Goat Anti-Rabbit IgG H&L (HRP) (ab97051) at 1/100000 dilution

      Predicted band size: 62 kDa
      Observed band size: 80 kDa
why is the actual band size different from the predicted?

The expression profile/ molecular weight observed is consistent with what has been described in the literature (PMID: 17974966).

      Negative control: HL-60 (PMID: 17974966).

      Blocking/Dilution buffer: 5% NFDM/TBST.

      Exposure times: Lanes 1-2: 60 seconds; Lane 3: 30 seconds; Lane 4: 10 seconds; Lane 5: 180 seconds.

    • Flow cytometric analysis of 4% paraformaldehyde-fixed, 90% methanol permeabilized HL-60 (human promyelocytic leukemia cell line, Left panel) HeLa (human cervix adenocarcinoma epithelial cell, Right panel) labeling Bag3 with ab209251 at 1/60 (red) compared with a Rabbit monoclonal IgG (ab172730) (black) and an unlabeled control (cells without incubation with primary antibody and secondary antibody) (blue). Goat anti rabbit IgG (Alexa Fluor® 488, ab150077), at 1/2000 dilution was used as the secondary antibody.

      Negative control: HL-60 (PMID: 17974966).
