    Does not react with: Zebrafish
  • Immunogen

Mix of cell preparations containing human cytokeratins.

  • General notes

Cytokeratins are a subfamily of intermediate filament proteins and are characterized by a remarkable biochemical diversity, represented in human epithelial tissues by at least 20 different polypeptides. They range in molecular weight between 40 kDa and 68 kDa and isoelectric pH between 4.9 – 7.8. The individual human cytokeratins are designated 1 to 20. The various epithelia in the human body usually express cytokeratins which are not only characteristic of the type of epithelium, but also related to the degree of maturation or differentiation within an epithelium. Cytokeratin subtype expression patterns are used to an increasing extent in the distinction of different types of epithelial malignancies. The cytokeratin antibodies are not only of assistance in the differential diagnosis of tumors using immunohistochemistry on tissue sections, but are also a useful tool in cytopathology and flow cytometric assays.

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

The cells were then incubated in 1x PBS / 10% normal goat serum / 0.3M glycine to block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab9222, 1µg/1x106 cells) for 30 min at 22ºC. The secondary antibody used was DyLight® 488 goat anti-mouse IgG (H+L) (ab96879) at 1/500 dilution for 30 min at 22ºC. Isotype control antibody (black line) was mouse IgG1 [ICIGG1] (ab91353, 2µg/1x106 cells) used under the same conditions. Acquisition of >5,000 events was performed. This antibody gave a positive signal in A431 cells fixed with 4% paraformaldehyde (10 min)/permeabilized with 0.1% PBS-Tween for 20 min used under the same conditions.

Question

I have a request/question. Do you have a positive control (protein sample) for keratin 5 that you could send me?

Answer

Thank you for your reply. I just checked our catalogue and unfortunately we do not have any positive control recombinant cytokeratin 5 proteins. This monoclonal antibody stains basal cells in combined and stratified epithelial tissues. It recognizes the stem cell population, including the so-called amplifying cells in the prostate epithelium. A good positive control may therefore be human prostate lysate like ab30304 or even skin like ab30166, but this is something that your lab would have to purchase. https://www.abcam.com/Prostate-Human-Tissue-Lysate-adult-normal-tissue-ab30304.html https://www.abcam.com/Skin-Human-Tissue-Lysate-adult-normal-tissue-ab30166.html I hope this information helps.
