  • Immunostaining of HEK 293 cells transfected with a human BCMA expression plasmid (left panel), or mock transfected (right panel).
    Method: 3 days after transfection of cells with the indicated constructs, cells were fixed with acetone or 4% formaldehyde for 5min at RT. Slides were blocked with normal IgG, and incubated for 1 hour with 5µg/ml ab17323 in 1%BSA / 1x PBS. After washes in PBS, samples were incubated with the secondary antibody for 1 hour, washed in PBS and revealed with StreptABComplex/HRP (Vector) and AEC.

  • FACS analysis of cells with ab17323 to BCMA.

    Method: HEK 293 cells were mock transfected or transfected with an expression plasmid coding for human BCMA. Cells (5x105) were incubated on ice for 30 min in 50µl FACS buffer (PBS, 5% fetal calf serum, 0.02% azide) containing 1µg/ml of ab17323 monoclonal antibody to human BCMA. After washing in FACS buffer, PE-conjugated antibody to rat IgG was added. Cells were incubated on ice for 30 min, washed and analyzed by flow cytometry.

  • Detection of endogenous human BCMA with ab17323 to BCMA (human)(Vicky-1) (second peak, to the right). Method: U266 cells (2x105) were incubated on ice for 30 minutes with 0.2 µg of ab17323 or an isotype control in 25µl FACS buffer (PBS, 5% Fetal calf serum, 0.02% azide). The primary antibody was revealed with anti- Rat IgG (R-PE) and then analyzed by Flow Cytometry.

Thank you for your enquiry. I am sorry to confirm that as far as we are aware, ab17323 has never been tested with samples from cynomolgus monkey. All tested and guaranteed species cross-reactivity information is stated on our datasheets, and these are updated as soon as any new information is brought to our attention. I can confirm the immunogen for this antibody is as follows: LQMAGQCSQ NEYFDSLLHA CIPCQLRCSS NTPPLTCQRY CNASVTNSVK GTNA   Unfortunately, I am sorry I am not able to find the cynomolgus monkey sequence available on the protein database sites. If you have access to this sequence, I can recommend to try an alignment. Or if you would like to send the sequence to me, I am happy to do this. We recommend that alignment should be over 85% to predict that an antibody will detect in a different species. I'm sorry to hear the customer is experiencing problems in FACS with some of our antibodies, it is likely that the problem is protocol related. I have received this morning a detailed FACS protocol with details of the buffers to use and fixative to use for staining with ab17323 and am still waiting for this information from the source of the anti TAC1 antibody, my apologies. The positive control recommended for endogenous detection by FACS is U266 cells. Recommended FACS protocol with ab17323 Buffers: PBS 1% Formaldehyde (FA) keep at 4°C PBS 0.5% saponin keep at 4°C PBS 50 mM NH4Cl 1.33g NH4Cl for 500ml Complete medium 0.5% saponin 3 x 105 cells/sample Protocol 1-wash cells 1X in PBS 2-incubate in 0.5 ml PBS 1%FA, 15 min at RT under agitation 3-wash 1X with PBS 4-wash 1X with PBS-NH4Cl 5-wash 1X with PBS 6-incubate with the first antibody in complete medium + 0.5% saponin for 30 min at RT. 7-wash once with 1X PBS 0.5% saponin 8-incubate with the secondary antibody in complete medium + 0.5% saponin for 30 min (or more depending on secondary specification) at RT in the dark. 9-wash once with PBS 0.5% saponin 10-wash once with PBS 11-resuspend in 0.2 ml PBS 12- analyse with FACS I hope this information helps,
