For BDNF, multiple WB bands are possible and expected. The human protein has 5 isoforms (precursors: 28 – 37 kDa) and can be glycosylated (Uniprot: P23560). The mature form is expected at ~14 kDa (monomer) and the dimer at ~28 kDa.

FunctionDuring development, promotes the survival and differentiation of selected neuronal populations of the peripheral and central nervous systems. Participates in axonal growth, pathfinding and in the modulation of dendritic growth and morphology. The versatility of BDNF is emphasized by its contribution to a range of adaptive neuronal responses including long-term potentiation (LTP), long-term depression (LTD), certain forms of short-term synaptic plasticity, as well as homeostatic regulation of intrinsic neuronal excitability.

Immunocytochemistry/ Immunofluorescence analysis of DIV9 rat E18 primary cortical neuron cells labeling BDNF with ab226843 at 1:500 (green). Cells were fixed in 4% paraformaldehyde at room temperature for 15 minutes. Cells were counterstained with DAPI (blue) and a Tau antibody diluted at 1/500 (yellow).
Immunohistochemical analysis of paraffin-embedded rat brain tissue labeling BDNF with ab226843 at 1/500 dilution at 4°C overnight. Antigen Retrieval:Citrate buffer, pH 6.0, 15 min (Cuisinart Electric Pressure Cooker #EPC-1200, choose "high pressure"). Endogenous peroxidase blocking: 3% H2O2, RT, 30min. Blocking:1.5% goat serum (dilute goat serum by 1xPBS), RT, 30 minutes. Secondary antibody incubation: ABC HRP Kit (Rabbit IgG), 1:200, RT, 30min. Washing: PBS, 2 x 5 mins. Chromogen system: DAB.
