    • Relevance

      Beta galactosidase is coded by a gene (lac z) in the lac operon of Escherichia coli. It is a metalloenzyme that splits lactose into glucose and galactose. It hydrolyzes terminal, non-reducing beta-D-galactose residues in beta-D-galactosides. Activation by cations seems to be substrate dependent. K+, Na+, NH4+, Rb+, Cs+ and Mn++ all activate enzyme activity based upon the substrate used.

      Inquiry: Dear customer service, I am really confused about this product. People are asking you if it could be suitable on human tissue sections and your answer is yes. Does it mean that this antibody cross reacts with human b-galactosidase? If so, does it also cross react with murine b-galactosidase? My understanding is that the immunogen is the E.coli form, therefore -unless the epitope is conserved between human and bacteria, it should not cross react. Many thanks for your reply

      I can confirm that ab4761 will detect beta Galactosidase from E.coli origin.
      If beta Galactosidase is overexpressed (ectopically expressed) in human tissue than this antibody is suitable for this.
      We tested this antibody with beta Galactosidase that was expressed in HeLa cells.

      The details you have kindly provided will enable us to investigate this case for you and this is also helpful in our records for monitoring of quality.
      I would like to reassure you that ab4761 is tested and covered by our 6 month guarantee for use in samples that overexpress beta Galactosidase and IHC. In the event that a product is not functioning in the applications cited on the product data sheet, we will be pleased to provide a credit note or free of charge replacement.
      Reviewing this case, I would like to offer some suggestions to help optimise the results from ab4761.
      1.) I recommend to use a permeabilization step since I assume that the beta Galactosidase will be overexpressed in the cytoplasm. 0.1% triton for 10 minutes only at RT.
      2.) The BSA in the primary antibody solution can be omitted.
      3.) I recommend to run a WB with the sample to confirm the expression of beta Galactosidase.
      4.) It might be worth to try without antigen retrieval.
      I would also appreciate if you can confirm some further details: 1.) Please give more details where the beta Galactosidase is overexpressed, what system was used and how the expression was tested.
      2.) Is the secondary antibody and the rest of the detection reagents working?

      I am a graduate student at Scottish Centre of Regenerative Medicine.

      We are interested in unconjugated rabbit or rat anti-beta-galactosidase antibody (anti-LacZ Ab). We plan to use this Ab on mouse embryonic tissues:routinelyfor whole mount staining andoccasionallyfor cryo-sections. We have found several anti-LacZ Abs on your web-site, but they seems are not recommended for our particular application.

      Those are ab616 and ab4761. ab616 is not recommended for immunohistochemistry according to yourdescription. The only option left is ab4761, however it was tested with parafin sections and WB; and there are no images. I have heard (on "Origins of tissue stem cells" conference organized with Abcam, June 29, 2012) that in such circumstances we can buy the antibody with some discount. Could you advise, please.

      We have several antibodies against beta galactosidase, two of which have been validated for IHC staining of paraffin embedded sections. You may want to consider chicken polyclonal ab9361 or rabbit polyclonal ab4761. These have not been validated in human tissue specifically but should be compatible in detecting the Beta galactosidase protein.
