  • General notes

The Life Science industry has been in the grips of a reproducibility crisis for a number of years. Abcam is leading the way in addressing the problem with our range of recombinant monoclonal antibodies and knockout edited cell lines for gold-standard validation.

One factor contributing to the crisis is the use of antibodies that are not suitable. This can lead to misleading results and the use of incorrect data informing project assumptions and direction. To help address this challenge, we have introduced an application and species grid on our primary antibody datasheets to make it easy to simplify identification of the right antibody for your needs.

Target

  • Function

The major proteolytic product p15 BID allows the release of cytochrome c (By similarity). Isoform 1, isoform 2 and isoform 4 induce ICE-like proteases and apoptosis. Isoform 3 does not induce apoptosis. Counters the protective effect of Bcl-2.
  • Tissue specificity

Isoform 2 and isoform 3 are expressed in spleen, bone marrow, cerebral and cerebellar cortex. Isoform 2 is expressed in spleen, pancreas and placenta (at protein level). Isoform 3 is expressed in lung, pancreas and spleen (at protein level). Isoform 4 is expressed in lung and pancreas (at protein level).
  • Domain

Intact BH3 motif is required by BIK, BID, BAK, BAD and BAX for their pro-apoptotic activity and for their interaction with anti-apoptotic members of the Bcl-2 family.
  • Post-translational
    modifications

    TNF-alpha induces a caspase-mediated cleavage of p22 BID into a major p15 and minor p13 and p11 products.
Phosphorylated upon DNA damage, probably by ATM or ATR.
p15 BID is ubiquitinated by ITCH; ubiquitination results in proteasome-dependent degradation.
  • Cellular localization

Cytoplasm; Cytoplasm. Mitochondrion membrane. When uncleaved, it is predominantly cytoplasmic; Mitochondrion membrane. A significant proportion of isoform 2 localizes to mitochondria, it may be cleaved constitutively; Mitochondrion membrane. Associated with the mitochondrial membrane and Mitochondrion membrane. Translocates to mitochondria as an integral membrane protein.

    Asked on Nov 10 2005

    Answer

Thank you for your enquiry. I have contacted the scientists that tested this antibody, and they stated that they only observe one single band at 22kDa with the antibody. I am sorry that we don't have any further insight as to what the 37kD band is that you are seeing. Please do not hesitate to contact us if you need anything further.

    Question

Thank you for your prompt answer. Could you find out if the peptide the company use as immunogen is in the portion where the proteins have 80 % homology with the rat or in the 20% where they are different. We already bought the antibody.I'm changing conditions in the western blot to see if I have the protein. We are working with a luteal cell line derived from rat.

    Answer

Thank you for your enquiry. All the information we have on species cross reactivity is specified on the datasheet. To our knowledge, cross reactivity with rat has not yet been tested. Should you decide to go ahead and purchase this product, please let us know how you get on and in return we will forward a reward of your choice, typically an Amazon gift voucher.
