RelevanceThe BID (BCL2 Interacting Domain) protein belongs to an important class of regulators of apoptosis. This BH3 domain containing proapoptotic Bcl2 family member, is localized in the cytosolic fraction of cells as an inactive precursor. Its active form is generated upon proteolytic cleavage by caspase 8 in the Fas signaling pathway. Cleaved Bid translocates to mitochondria and releases its potent proapoptotic activity, which in turn induces cytochrome c release and mitochondrial damage. The cytochrome c releasing activity of Bid was antagonized by Bcl2. Mutation in the SH3 domain can diminish the cytochrome c releasing activity. In animal model studies, Bid deficient mice are found resistant to the lethal effects of death factor signals relayed through Fas.

Lane 1 : NIH/3T3 whole cell lysate untreated at 50 µg
Lane 2 : NIH/3T3 whole cell lysate treated with Etoposide (100 µM , 3 hrs) at 50 µg
Lane 3 : NIH/3T3 whole cell lysate untreated at 80 µg
Lane 4 : NIH/3T3 whole cell lysate treated with Etoposide (100 µM , 3 hrs) at 80 µg
Predicted band size: 22 kDa
Exposure time: 3 minutes
Whole cell lysate (1 mg for IP, 20% of IP loaded) from NIH/3T3 cells treated with 100 μM etoposide for 3 hours (+) or mock treated (-) cells. ab264131 was used for WB at 1 μg/ml. An alternative Rabbit anti-BID antibody was used for IP for BID and phospho-BID.
