No secondary antibody used however, after primary antibody incubation, slides were washed and stained with Streptavidin-550 (DyLight) at 1:500 dilution for 30-60 minutes at room temperature. Perfusion is not necessary. Use 4% PFA over night. *For bone sections, decalcification in 14% EDTA for 24-72 hours. *Cryopreserve in 30% sucrose for 24-72 hours. Over night primary antibody incubation is not necessary (alternative: room temperature for 1 hr.).
