      Component of the class I major histocompatibility complex (MHC). Involved in the presentation of peptide antigens to the immune system.
    • Involvement in disease

      Defects in B2M are the cause of hypercatabolic hypoproteinemia (HYCATHYP) [MIM:241600]. Affected individuals show marked reduction in serum concentrations of immunoglobulin and albumin, probably due to rapid degradation.
      Note=Beta-2-microglobulin may adopt the fibrillar configuration of amyloid in certain pathologic states. The capacity to assemble into amyloid fibrils is concentration dependent. Persistently high beta(2)-microglobulin serum levels lead to amyloidosis in patients on long-term hemodialysis.

        IHC image of beta 2 Microglobulin staining in human normal lung formalin fixed paraffin embedded tissue section, performed on a Leica BondTM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab21899, 10µg/ml, for 15 mins at room temperature and detected using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

        For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

      When using ab21899 Anti-beta 2 Microglobulin antibody [B2M-01] (Biotin) in IP we can recommend a working range of 1-10 ug/ml. The exact concentration for your IP experiments should be within this range but could be outside of this and will need to be determined experimentally.

      Abcam also provides a number of products for use in IP such as our https://www.abcam.com/ab119211 ready to use protein gel stain, our https://www.abcam.com/index.html?pageconfig=resource&rid=15115#1 and our https://www.abcam.com/index.html?pageconfig=resource&rid=15115#2 . Our VeriBlot line only recognize native (non-reduced) antibodies and are recommended when the target of interest is <30kDa, whereas our Light Chain specific line do not recognize heavy-chains and are recommended when your protein of interest is >30kDa.       Question

      Yes, I want to coat the plate with ab94886 in order to captureHLA-G onto the plate. Then I want to detect beta 2 microglobulin(because both proteins are ensemble together in the native form) withab21889.
      So, if I want to detect these two proteins, are these clones I chose the best for ELISA? (Specially for beta 2 microglobulin, as I saw there are two distinct clones both for ELISA)

      These two monoclonals may be an effective pair but they have not been tested with each other in sandwich ELISA. My concern is whether or not the epitope (the binding site) that the beta 2 microglobulin antibody ab21899 recognizes is accessible if it is in an ensemble/complex with the HLA G. That is also the case for the other monoclonal, ab25160.

      A better choice may be a polyclonal antibody raised against full-length beta 2 microglobulin, which will recognize many epitopes, not just one. I suggest ab34678, a biotinylated rabbit polyclonal.

      However, this has not been tested in sandwich ELISA with ab94886 either. We do not have any pairs that have been validated for sandwich ELISA but ab94886 and ab34678 are my recommendation. (Please note that these have only been tested against human samples).
