  • Relevance

    CCL6 (C10) is a small cytokine belonging to the CC chemokine family that has only been identified in rodents. It is a potent chemoattractant of macrophages, but it can also attract B cells, CD4+ lymphocytes and eosinophils. In mice, CCL6 is expressed in cells from neutrophil and macrophage lineages, and can be induced under conditions suitable for myeloid cell differentiation. CCL6 (C10) was originally identified as a transcript that is induced in bone marrow cells upon stimulation of GMCSF. The cell surface receptor for CCL6 is believed to be the chemokine receptor CCR1.

    Question

    My question is whether the "highly pure recombinant mouse CCL6" you used to immunize the animals is:
    (1) complete full length protein containing N-terminal signal peptide (a.a 1-21)?,
    (2) secreted protein lacking signal peptide but maintain whole other parts (a.a 22-116)? or
    (3) mature active protein lacking the N-terminal domain (22-43), which contains only a.a. 44-116?
    I can agree with your statement that epitope mapping is usually not performed for validating poly-clonal antibodies.
    So the details of the immunogen is quite important to estimate the reactivity of the antibody.
    If (3) above is the case, we don't have to worry about the reactivity because this antibody must react with all forms of CCL6.
    But if (1) or (2) is the case, it might happen that this antibody preferentially react with inactive form of CCL6 with N-terminal a.a. 1-43 or 22-43.
    So if possible, please give me the details of recombinant CCL6 used as an immunogen.

    The recombinant MurineCCL6 (C10) is also in our catalogue ab9898. This is the mature protein, which contains 95 amino acids (amino acids 22-116).

    The amino acid sequence is as follows:

    GLIQEIEKED RRYNPPIIHQ GFQDTSSDCC FSYATQIPCK RFIYYFPTSG GCIKPGIIFI SRRGTQVCAD PSDRRVQRCL STLKQGPRSG NKVIA

    The 21 amino acid N-terminal signal peptide is not present on the protein.

    Lot number: GR82949-1
    Inquiry: We purchased the antibody above (anti-CCL6 biotin) for WB, and could see some signals in our samples using this antibody. But the band size we saw was significantly larger than the expected. Since CCL6 is processed from its inactive form (pro-peptide) to the active mature form, we assume that the band we saw was the inactive pro-peptide. We want to know if the antibody could also detect the active form of CCL6, to know if our samples contain active CCL6. For this, we ask two specific questions: (1) The immunogen used to raise this antibody is the active (truncated) CCL6 or full-length inactive CCL6? (2) If full length you used, have you confirmed epitopes this antibody recognizes? Many thanks

    Thank you for your enquiry regarding ab83134.

    The immunogen used to raise this antibody was a highly pure (>98%) recombinant mouse CCL6. This is a polyclonal antibody and will therefore consist of a mixed population of IgGs each of which will recognize a different epitope. For polyclonal antibodies epitope mapping experiment is not carried out.

    Currently, as our datasheet indicates we have no data with this product on cell/tissue samples yet this needs to be tested.
