Target

  • Function

    Ghrelin is the ligand for growth hormone secretagogue receptor type 1 (GHSR). Induces the release of growth hormone from the pituitary. Has an appetite-stimulating effect, induces adiposity and stimulates gastric acid secretion. Involved in growth regulation.
    Obestatin may be the ligand for GPR39. May have an appetite-reducing effect resulting in decreased food intake. May reduce gastric emptying activity and jejunal motility.
  • Tissue specificity

    Highest level in stomach. All forms are found in serum as well. Other tissues compensate for the loss of ghrelin synthesis in the stomach following gastrectomy.
  • Sequence similarities

    Belongs to the motilin family.
  • Post-translational
    modifications

    O-octanoylation or O-decanoylation is essential for ghrelin activity. The O-decanoylated forms Ghrelin-27-C10 and Ghrelin-28-C10 differ in the length of the carbon backbone of the carboxylic acid bound to Ser-26. A small fraction of ghrelin, ghrelin-28-C10:1, may be modified with a singly unsaturated carboxylic acid.
    Amidation of Leu-98 is essential for obestatin activity.

    Question

    The antibody ab48285 at 4 ug/ml did not work well in our system. In fact, by using this much antibody, we can only make 25 ml of 4 ug/ml. In each RIA tube, we have the incubation volume of 0.5 ml. Thus, with 100 ug of ab48285 we can only run 50 tubes. If we would run ab48285 at 8 ug/ml, then we can only run 25 tubes. As you can see that this would not be economical. When we ordered this antibody, we were hoping that it would work out at 2 ug/ml; we tested both 2 ug/ml and 4 ug/ml, they yielded 5% and 10 % specific binding, respectively. With this kind of binding result, I do not expect 8ug/ml of ab 48285 will give us adequate binding for RIA.

    I hope you will give us credit at this time.

    We had good experience with your products, and will not hesitate to try more of your products in the future.

    Question

    I was testing your product rabbit polyclonal to ghrelin (ab48285) in our RIA system, and it failed. Following your instruction, I used 4 ug/ml but the specific binding of 125I-ghrelin was only 10%, whereas the antibody from another company showed 30% specific binding. I understand that you would refund the cost if the customer is not satisfied with your product.

    I am sorry that you have been experiencing difficulties with this antibody in your RIA experiments. I understand that you have tested a concentration of 4ug/ml - have you tested any others? We recommend up to 8ug/ml. Some further optimization may be required for best results with this antibody.

    The antibody isrecommended from 2-8 ug/ml for RIA purpose. The efficiency of labeling of their tracer is another factor to determine the concentration use for their RIA assay.The optimal condition would need to be determined by the enduser.

Anti Orexin ab35337, ab6214, ab16787, ab64565 These antibodies are tested in Indirect ELISA so are guaranteed for this application and applications as stated on the datasheets. Anti leptin ab20101, ab20115, ab9749 and ab2095 Anti Insulin ab6995, ab63820, ab7760 and ab46707 Anti neuropeptide Y We ab30914, ab10341, ab6173, ab43871, ab53478. These antibodies are not yet tested in ELISA. Anti peptide YY  ab51537, ab51539 etc. Anti ACTH  ab20358, ab21003, ab104768,a b104619 Anti MCH ab37337; These are however not yet tested in ELISA.
