Target

  • Function

Plasmin dissolves the fibrin of blood clots and acts as a proteolytic factor in a variety of other processes including embryonic development, tissue remodeling, tumor invasion, and inflammation. In ovulation, weakens the walls of the Graafian follicle. It activates the urokinase-type plasminogen activator, collagenases and several complement zymogens, such as C1 and C5. Cleavage of fibronectin and laminin leads to cell detachment and apoptosis. Also cleaves fibrin, thrombospondin and von Willebrand factor. Its role in tissue remodeling and tumor invasion may be modulated by CSPG4. Binds to cells.
Angiostatin is an angiogenesis inhibitor that blocks neovascularization and growth of experimental primary and metastatic tumors in vivo.
  • Tissue specificity

Present in plasma and many other extracellular fluids. It is synthesized in the liver.
  • Involvement in disease

    Plasminogen deficiency
  • Sequence similarities

Belongs to the peptidase S1 family. Plasminogen subfamily.
    Contains 5 kringle domains.
    Contains 1 PAN domain.
    Contains 1 peptidase S1 domain.
  • Domain

Kringle domains mediate interaction with CSPG4.
  • Post-translational
    modifications

N-linked glycan contains N-acetyllactosamine and sialic acid. O-linked glycans consist of Gal-GalNAc disaccharide modified with up to 2 sialic acid residues (microheterogeneity).
In the presence of the inhibitor, the activation involves only cleavage after Arg-580, yielding two chains held together by two disulfide bonds. In the absence of the inhibitor, the activation involves additionally the removal of the activation peptide.
  • Cellular localization

Secreted. Locates to the cell surface where it is proteolytically cleaved to produce the active plasmin. Interaction with HRG tethers it to the cell surface.

    Question

Dear Sir or Madam, I am looking for a blocking antibody that would specifically inhibit human plasmin activity. I was not able to find one on your website but I may be wrong : do you indeed sell such antibodies? I would be very grateful if you would consider my request, I am looking from hearing from you at your earliest convenience, Yours sincerely,

    Answer

    Thank you for contacting us.

We currently offer only one anti-Plasmin antibody ab48350. This product has not been tested as a neutralizing/blocking antibody for plasmin. As a rabbit polyclonal this product will recognize a number of epitopes on plasmin and therefore may neutralize it, however there will also be lot to lot variation in epitope recognition.

For UNTESTED species and/or applications, we have established a testing discount program. Here is a brief description of how it works:

The testing discount program is for customers who like to use an antibody/protein on an untested species/application. You would purchase the antibody at full price, test it and submit an Abreview with your data (positive or negative). On your next order you will receive a discount for ONE antibody at the full price (100%) of the antibody you have tested. The terms and conditions applicable to this offer can be found here: https://www.abcam.com/collaborationdiscount.

I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.

    Question

    - about specifics of this antibody - why anti plasmin and not plasminogen?
    - has this antibody been tested against plasmin when complexed to PNA or PAI?

    Answer

    Question

I have further questions regarding this product. Does this antibody only react to plasmin? Or does it also react to plasminogen? When you confirmed that this antibody only reacts to mouse plasmin 15%, does that mean the remainder 85% reacts to plasminogen? There is no amino acid difference between plasmin and plasminogen. Although antibodies are named plasmin, most of them will also react to plasminogen. Can you please confirm if this antibody will react to plasminogen?

    Answer

Thank you for your enquiry. This antibody has not been tested if they can recognize plasminogen. Therefore, I can not confirm if it only detects plasmin. This antibody reacts 15% to mouse sample, which means that the remaining 85% is no reaction.
