    I am sorry to confirm that as far as we are aware, ab77310 has never been tested in sandwich ELISA (sELISA). It has been tested in indirect ELISA only (ELISA). All tested and guaranteed applications are specified on our datasheets, and these are updated as soon as any new information is brought to our attention.

    Sandwich ELISA procedures can be difficult to optimize and tested match pair antibodies should be used. This ensures the antibodies are detecting different epitopes on the target so they do not interfere with the other antibody binding. Therefore, I regrettably we are unable to guarantee our antibodies in sandwich ELISA unless they have been specifically tested for sandwich ELISA.

    Reviewing the details on the datasheet forab10184 Anti-Prostate Specific Antigen antibody [2H9], this has been tested in sELISA. Several matched pairs are suggested for use with this antibody:

    'Can be paired for Sandwich ELISA with Mouse monoclonal [5A6] to Prostate Specific Antigen (ab10185) and Mouse monoclonal [8A6] to Prostate Specific Antigen (ab10187) and Mouse monoclonal [5A6] to Prostate Specific Antigen (HRP) (ab24466).

    For the detection of total PSA, ab10184 can be used as a capture antibody in conjugation with ab10185 or with ab24466 (HRP-conjugated version). For the detection of free PSA, ab10184 can be used as a capture antibody in conjugation with ab10187.'

    I can recommend to review the online datasheets for further information. I suggest it would be more appropriate for the customer to use one of these matched pairs suggested above with ab10184which we can guarantee for sandwich ELISA:

    I have reviewed the details of this experiment and have the following feedback:

    1. ab119154 Prostate Specific Antigen protein is full length human protein, so should be detected by both ab77310 and ab102885.

    2. The ab102885 Easylink conjugation kit would be suitable for conjugating FITC to ab10187.

    3. In theory, the experiment the customer has described should work. However, these antibodies have not been tested in this type of application, an I am sorry we would not be able to guarantee them for this.

    4. Also, as discussed in the previous email, for sandwich ELISa type experiments, we would suggest to use a matched pair. ab10187 has been tested as a matched pair withthe following:

    Sandwich ELISA (sELISA) procedures can be difficult to optimize and tested match pair antibodies should be used. Any tested matched pairs would be noted on the datasheet. This ensures the antibodies are detecting different epitopes on the target protein so they do not interfere with the other antibody binding. Therefore, I am sorry we are unable to guarantee our antibodies in sandwich ELISA unless they have been specifically tested for sELISA.

    The Prostate Specific Antigen antibody [2H9] ab10184 that your customer has found has been tested in sandwich ELISA.The information on the datasheet states that this can be paired for sELISA with one of the the following:

The whole blood will first be enriched for prostate cancer cells via  a microfluidic device capturing via a mAb for a specific cell receptor. 3.
